Plants adapt to their changing environments by sensing and responding to physical, biological, and chemical stimuli. Due to their sessile lifestyles, plants experience a vast array of external stimuli and selectively perceive and respond to specific signals. By repurposing the logic circuitry and biological and molecular components used by plants in nature, genetically encoded plant-based biosensors (GEPBs) have been developed by directing signal recognition mechanisms into carefully assembled outcomes that are easily detected. GEPBs allow for monitoring of biological processes in plants to facilitate basic studies of plant growth and development. GEPBs are also useful for environmental monitoring, plant abiotic and biotic stress management, and accelerating design-build-test-learn cycles of plant bioengineering. With the advent of synthetic biology, biological and molecular components derived from alternate natural organisms (e.g., microbes) and/or parts have been used to build GEPBs. In this review, we summarize the framework for engineering different types of GEPBs. We then highlight representative validated biological components for building plant-based biosensors, along with various applications of plant-based biosensors in basic and applied plant science research. Finally, we discuss challenges and strategies for the identification and design of biological components for plant-based biosensors.

Plants have evolved sophisticated molecular switches to perceive and respond to both endogenous and exogenous signals, enabling changes in form and function in response to environmental and developmental cues. As autotrophic, multicellular organisms, plants represent promising platforms for designing and engineering sense-and-report modules. Advances in protein engineering and functional screening have facilitated the reprogramming of native switches into biosensors capable of detecting novel small molecules.

Plants as living organisms, as well as their material-structural components and physiological processes, offer promising elements for developing more sustainable technologies. Previously, we demonstrated that plants could acquire electronic functionality, as their enzymatic activity catalyzes the in vivo polymerization of water-soluble conjugated oligomers. We then leveraged plant-integrated conductors to develop biohybrid energy storage devices and circuits.

This manuscript reports the application of sensors for water use efficiency with a focus on the application of an in vivo OECT biosensor. In two distinct experimental trials, the in vivo sensor bioristor was applied in yellow kiwi plants to monitor, in real-time and continuously, the changes in the composition and concentration of the plant sap in an open field during plant growth and development. The bioristor response and physiological data, together with other fruit sensor monitoring data, were acquired and combined in both trials, giving a complete picture of the biosphere conditions.
