In the present work, heterocyclic compounds containing different moieties, such as pyrazole and thiophene, were synthesized and screened for inhibitory potency against medicinal enzymes and bacterial and cancer (breast and cervical) cell lines. The synthesized compounds have exhibited inhibitory capability against the studied enzymes. Among substances, compound showed AChE and BChE inhibitory potency with the lowest IC value of 3.72 ± 0.57 and 1.66 ± 0.22 µM, respectively, in comparison to the standard tacrine. These analogs indicated varying degrees of tyrosinase inhibitory potencies ranging from 1.12 ± 0.50 to 7.70 ± 0.88 µM, and substance was more potent against the enzyme than the reference compound, kojic acid. All four compounds have IC values between 37.11 ± 1.56-124.8 ± 2.09 µM for α-glucosidase. It was found that compound exhibited a better antiproliferative activity compared to other substances, with IC values at 5.068 and 6.460 µg mL for MCF-7 and HeLa cells, respectively. and compounds had good inhibitory ability against with a MIC value (16 µg mL-ˡ). Molecular docking analysis showed that C3 has the lowest binding score against α-glucosidase (-8.617 kcal/mol).Communicated by Ramaswamy H. Sarma.

The present study aimed to unveil the gastroprotective potential of Vaccinium macrocarpon (VM) extract and its mechanism of action against indomethacin (INDO)-induced gastric ulcers in rats. To achieve this goal, rats were pretreated with either omeprazole (20 mg/kg) or VM (100 mg/kg) orally for 14 consecutive days. Gastric tissue samples were collected and various parameters were evaluated to understand the mechanism of VM's action, including the levels of superoxide dismutase, malondialdehyde, glutathione, CAT and transforming growth factor beta (TGF-β), as well as the mRNA expression levels of tumour necrosis factor alpha, interleukin 1 beta, nuclear factor kappa B (NF-κB) and inhibitor kappa B (IκB).

Purpose Of Review: Mast cell activation is defined by activation of mast cells by varying stimuli with release of chemical mediators either through degranulation or release of de novo synthesized proteins or lipid mediators. Currently, tryptase measurement increase during symptomatic episodes is the most accepted biomarker measurement for mast cell activation. However, newer diagnostic tools including clinically available urinary mast cell mediators are noninvasive and can be more readily obtained compared to serum tryptase levels.

This study was aimed to identify the most frequently observed pathogens in uncomplicated urinary tract infections from outpatient urinary isolates obtained across seven different geographical regions in Türkiye and to determine whether the antibiotic resistance rates of these pathogens differ significantly between these regions. The study included patients aged 18 to 65 years who were diagnosed with uncomplicated urinary tract infections and had positive urine cultures from March 2021 to August 2022, across 37 different centers in Türkiye. The participating centers were selected based on their use of the disk diffusion method, in line with the European Committee on Antimicrobial Susceptibility Testing (EUCAST) guidelines, to ensure standardization of urine culture data.
