Kinetic assays with recombinant enzymes are critical to determine the steady state kinetic parameters for androgen conversion to understand their role in androgen biosynthesis and metabolism. Detection and quantification of 5α-reduced androgens remain difficult to assay because they are UV-transparent compounds. Therefore, radioactive isotopic versions of these compounds are often required to conduct steady-state kinetics. Here we developed a derivatization protocol with dinitrophenylhydrazine (DNPH) to form hydrazones on the ketones of androgens enabling them to be detected by UV-reverse phase high performance liquid chromatography (RP-HPLC). We determined the kinetic parameters for the conversion of 5α-androstane-3,17-dione (5AD) to 5α-dihydrotestosterone (DHT), 11-keto-5α-androstane-3,17-dione (11K-5AD) to 11-keto-5α-dihydrotestosterone (11K-DHT), and 11β-hydroxy-5α-androstane-3,17-dione (11β-OH-5AD) to 11β-hydroxy-5α-dihydrotestosterone (11β-OH-DHT) catalyzed by recombinant aldo-keto reductase 1C3 (AKR1C3) as measured by product formation post DNPH derivatization.

Enzyme-instructed signal generation at liquid-liquid interfaces presents a novel strategy for controlling and detecting biochemical processes on macroscopic scales. Here, we explore the self-assembly and jamming of pillar[5]arene (P[5]A) derivatives at the oil-water interface via a copper-mediated "click" reaction, providing a versatile platform for generating observable signals. The formation of a pillar[5]arenes network at the droplet interface reduces interfacial tension, allowing droplets to adopt various nonequilibrium shapes based on the interfacial jamming process.

These herbicides give rise to a number of key metabolites, with 3-methyl phosphinicopropionic acid (MPPA), -acetyl glufosinate (-acetyl GLUF), aminomethyl phosphonic acid (AMPA), -acetyl aminomethyl phosphonic acid (-acetyl AMPA), -acetyl glyphosate (-acetyl GLY), -methyl glyphosate (-methyl GLY) as the major metabolites obtained from GLUF and GLY. Extensive use of these herbicides may lead to their increased presence in the environment, especially aquatic ecosystems.

Thromboxane A (TXA), a prothrombotic factor that induces platelet aggregation and thrombosis, acts as a vasoconstrictor by activating TXA receptors (TP receptors). TXA is extremely unstable and metabolizes into three major metabolites: 2,3-dinor thromboxane B (2,3-dinor-TXB), 11-dehydro TXB(11-dh-TXB), and 11-dehydro-2,3-dinor TXB(11-dh-2,3-dinor-TXB). 8-Iso-prostaglandin F(8-iso-PGF), a prostaglandin-like compound widely considered the best biomarker of oxidative stress, can also activate TP receptors.
