Hydrogen-bonded organic frameworks (HOFs) are under fast development in broad applications but have not been well explored for chemiresistive gas sensing yet primarily due to insufficient active sites. Herein, a new porphyrin-based HOF-199 is constructed by OH···O hydrogen bonds featuring layered networks and rich free oxygen (O) atoms, which is further exfoliated into few-layer nonosheets with more dangling O sites through an ultrasound-assisted liquid exfoliation method (namely L-HOF-199). Benefiting from rich electron-donor sites, L-HOF-199 demonstrates exceptional NO sensing properties under ambient conditions, achieving a remarkable 3.

Cell-free systems are powerful synthetic biology technologies that can recapitulate gene expression and sensing without the complications of living cells. Cell-free systems can perform more advanced functions when genetic circuits are incorporated. Here we expand cell-free biosensing by engineering a highly specific isothermal amplification circuit called polymerase strand recycling (PSR), which leverages T7 RNA polymerase off-target transcription to recycle nucleic acid inputs within DNA strand displacement circuits.

An innovative colorimetric sensing strategy was developed for the detection of glucose by the integration of glucose aptamer, glucose oxidase (GOx), and horseradish peroxidase (HRP), termed aptamer proximal enzyme cascade reactions (APECR). In the presence of glucose, aptamer binding enables GOx to catalyze glucose oxidation into HO efficiently. Subsequently, the adjacent HRP catalyzes the oxidation of the peroxidase substrate, 2,2'-biazobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), utilizing the generated HO, resulting in a distinct color change.
