The incorporation of fluorinated groups into peptides significantly affects their biophysical properties. We report herein the synthesis of Fmoc-protected trifluoromethylthiolated tyrosine (CFS-Tyr) and tryptophan (CFS-Trp) analogues on a gram scale (77-93% yield) and demonstrate their use as highly hydrophobic fluorinated building blocks for peptide chemistry. The developed methodology was successfully applied to the late-stage regioselective trifluoromethylthiolation of Trp residues in short peptides (66-80% yield) and the synthesis of various CFS-analogues of biologically active monoamines. To prove the concept, Fmoc-(CFS)Tyr and -Trp were incorporated into the endomorphin-1 chain () and into model tripeptides by solid-phase peptide synthesis. A remarkable enhancement of the local hydrophobicity of the trifluoromethylthiolated peptides was quantified by the chromatographic hydrophobicity index determination method, demonstrating the high potential of CFS-containing amino acids for the rational design of bioactive peptides.

Monoterpenes (MTs), the major constituents of plant essential oils, cover a broad spectrum of biological activities through their interaction with biomembranes. MTs are highly hydrophobic substances with a net electrical dipole, but are not clearly amphipathic. As a result, they aggregate at increasing concentrations in aqueous media, and in membrane environments their behavior changes from dynamics modulators to disruptors.

Hydrophobins are small amphiphilic proteins that confer filamentous fungal hydrophobicity needed for hyphal growth, development, dispersal and adhesion to host and substrata. In insect-pathogenic Beauveria bassiana, nine hydrophobins (class I Hyd1A-F and class II Hyd2A-C) were proven to localize on the cell walls of aerial hyphae and conidia but accumulate in the vacuoles and vesicles of submerged hyphae and blastospores, respectively. Conidial hydrophobicity, adhesion to insect cuticle, virulence via normal cuticle infection and dispersal potential were significantly more reduced by the hyd1A deletion leading to complete ablation of slender rodlets on conidial coat than the hyd1B deletion, which caused a failure to assemble morphologically irregular rodlets into orderly bundles.

Bicelles, an artificial disk-shaped lipid bilayer, are commonly used for the structural and functional characterization of membrane-bound proteins in an environment similar to that in intracellular membranes. Because the dynamics of the lipids that constitute bicelles exert a significant impact on the structure and function of the inserted proteins, we investigated the mobility of lipid molecules in bicelles composed of DMPC (14:0 PC) and DHPC (06:0 PC) using solution NMR and MD calculations. C R relaxation experiments for the acyl groups demonstrated that increasing bicelle sizes limit the rotational diffusion of acyl chain H-C bonds in DMPC.

Despite the importance of the effect of subnanoscale roughness on contact line behavior, it is difficult to directly observe the local behavior of contact lines at the micro- and nanoscale, leaving significant gaps in our current understanding. In this research, we investigate contact line motions and their relationship with nanoscale surface topography using coherence scanning interferometry. Our experiments were conducted on the substrates with different wettability without changing nanoscale surface topography.
