There is still a large amount of ferulic acid (FA), an outstanding antioxidant, present in agricultural residues. Enzymatic hydrolysis has been regarded as the most effective way to release FA. This present study therefore selected feruloyl esterase (FAE) and xylanase (XYN) from the metagenomes of a cow rumen and a camel rumen, respectively, for their recombinant expression in BL21 and further application in releasing FA. After screening the candidate signal peptides, the optimal one for each enzyme, which were selected as SP1 and SP4, respectively, was integrated into the vectors pET22b(+) and pETDuet-1. Among the generated strains SP1-F, SP4-X, and SP1-F-SP4-X that could express extracellular enzymes either separately or simultaneously, the latter one performed the best in relation to degrading the biomass and releasing FA. Under the optimized culture and induction conditions, the strain SP1-F-SP4-X released 90% of FA from 10% of de-starched wheat bran and produced 314.1 mg/L FA, which was deemed to be the highest obtained value to the best of our knowledge. This result could pave a way for the re-utilization of agricultural residues and enhancing their add-value.

G protein-coupled receptor 4 (GPR4) belongs to the subfamily of proton-sensing GPCRs (psGPCRs), which detect pH changes in extracellular environment and regulate diverse physiological responses. GPR4 was found to be overactivated in acidic tumor microenvironment as well as inflammation sites, with a triad of acidic residues within the transmembrane domain identified as crucial for proton sensing. However, the 3D structure remains unknown, and the roles of other conserved residues within psGPCRs are not well understood.

Propyrisulfuron, a novel sulfonylurea herbicide, effectively suppresses intracellular acetolactate synthase activity for weed control, but its adsorption behavior in the soil environment remains unclear. To assess potential agroecosystem risks, the adsorption-desorption behavior and mechanism of propyrisulfuron in six typical agricultural soils of China were investigated using a batch equilibrium method, Density Functional Theory (DFT), Fourier Transform Infrared Spectroscopy (FTIR), and Scanning Electron Microscopy equipped with Energy Dispersive X-ray (SEM-EDX) techniques. It is indicated that the adsorption-desorption of propyrisulfuron in six soils reached equilibrium at 36 hours under the optimum water-to-soil ratio (WSr) of 5:1.

This study introduces an innovative electrochemical biosensor, engineered through the functionalization screen-printed electrode (SPE) with a coordination complex comprised of 4-mercaptobenzoic acid (4-MBA) and copper ions (Cu), achieving precise quantitative determination of glyphosate. Electrodepositing gold nanoparticles (AuNPs) onto the electrode surface, forming a self-assembled monolayer (SAM) of 4-MBA via thiol-gold interactions, and immobilizing Cu via coordination bonding with the monolayer, finalizing the electrochemical biosensor construction as Cu/4-MBA/AuNPs/SPE. The successful modification of the biosensor interface is confirmed through scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), and electrochemical characterization.
