  • Celery seeds are effective in managing hyperuricemia and reducing gout recurrence by targeting xanthine oxidase (XOD), the enzyme that produces uric acid.
  • Researchers used a method called UF-LC-MS to quickly identify the compounds in celery seed extracts that have XOD-inhibitory effects.
  • They discovered 32 compounds, including several flavonoids, and confirmed that certain compounds can significantly inhibit XOD activity, establishing a chemical basis for celery seeds as an anti-gout supplement.

Article Abstract

Celery seeds have been used as an effective dietary supplement to manage hyperuricemia and diminish gout recurrence. Xanthine oxidase (XOD), the critical enzyme responsible for uric acid production, represents the most promising target for anti-hyperuricemia in clinical practice. In this study, we aimed to establish a method based on affinity ultrafiltration-liquid chromatography-mass spectrometry (UF-LC-MS) to directly and rapidly identify the bioactive compounds contributing to the XOD-inhibitory effects of celery seed crude extracts. Chemical profiling of celery seed extracts was performed using UPLC-TOF/MS. The structure was elucidated by matching the multistage fragment ion data to the database and publications of high-resolution natural product mass spectrometry. Thirty-two compounds, including fourteen flavonoids and six phenylpeptides, were identified from celery seed extracts. UF-LC-MS showed that luteolin-7--apinosyl glucoside, luteolin-7--glucoside, luteolin-7--malonyl apinoside, luteolin-7--6'-malonyl glucoside, luteolin, apigenin, and chrysoeriol were potential binding compounds of XOD. A further enzyme activity assay demonstrated that celery seed extract (IC = 1.98 mg/mL), luteolin-7--apinosyl glucoside (IC = 3140.51 μmol/L), luteolin-7--glucoside (IC = 975.83 μmol/L), luteolin-7--6'-malonyl glucoside (IC = 2018.37 μmol/L), luteolin (IC = 69.23 μmol/L), apigenin (IC = 92.56 μmol/L), and chrysoeriol (IC = 40.52 μmol/L) could dose-dependently inhibit XOD activities. This study highlighted UF-LC-MS as a useful platform for screening novel XOD inhibitors and revealed the chemical basis of celery seed as an anti-gout dietary supplement.

Introduction: Dl-3-n-butylphthalide (NBP), a small molecular compound extracted from celery seeds, has been shown to exhibit diverse pharmacological activities, including anti-inflammatory, antioxidative, and anti-apoptotic effects. Recent studies have highlighted its efficacy in treating various cardiovascular conditions, such as myocardial infarction, hypertrophy, heart failure, and cardiotoxicity. This study aimed to investigate whether NBP could alleviate cardiac dysfunction and injury following hemorrhage-induced cardiac arrest (HCA) in a porcine model and elucidate its potential mechanisms.

Article Synopsis
  • Drought significantly affects plant yield and quality, and this study examines how melatonin (MT) helps improve celery seedlings' resilience to drought stress.
  • The results showed that applying exogenous MT reduced damage to the seedlings, improved various physiological parameters, and increased overall drought tolerance by positively affecting chlorophyll production and energy conversion.
  • Transcriptomic analysis indicated that MT regulates important genes related to chlorophyll synthesis and glycolytic pathways, enhancing pyruvate accumulation and peroxidase activity, which are crucial for drought resistance in plants.
