Effect of betanin synthesis on photosynthesis and tyrosine metabolism in transgenic carrot.

Background: Betalain is a natural pigment with important nutritional value and broad application prospects. Previously, we produced betanin biosynthesis transgenic carrots via expressing optimized genes CYP76AD1S, cDOPA5GTS and DODA1S. Betanin can accumulate throughout the whole transgenic carrots. But the effects of betanin accumulation on the metabolism of transgenic plants and whether it produces unexpected effects are still unclear.

Results: The accumulation of betanin in leaves can significantly improve its antioxidant capacity and induce a decrease of chlorophyll content. Transcriptome and metabolomics analysis showed that 14.0% of genes and 33.1% of metabolites were significantly different, and metabolic pathways related to photosynthesis and tyrosine metabolism were markedly altered. Combined analysis showed that phenylpropane biosynthesis pathway significantly enriched the differentially expressed genes and significantly altered metabolites.

Conclusions: Results showed that the metabolic status was significantly altered between transgenic and non-transgenic carrots, especially the photosynthesis and tyrosine metabolism. The extra consumption of tyrosine and accumulation of betanin might be the leading causes.

Methods: This study aims to use field trials, starting from physiological responses, combined with proteomics and untargeted metabolomics, to systematically reveal the physiological and biochemical characteristics and key molecular mechanisms of tobacco leaves under chilling stress.

In order to make use of the large biomass of tomato plant to fulfill the purpose of remediating-while-producing, two commercial tomato varieties, 'Baiguoqiangfeng' (BG) and 'Ouguan' (OG) were grown in Cd contaminated acidic soil to compare their performance on Cd phytoextraction, and monocalcium phosphate (Ca) was foliar applied to reduce their fruit Cd concentration. The results showed that the BG was a more Cd tolerant variety, comparing with OG, it suffered lighter tissue peroxidation and photosynthesis obstacle, owning weaker amino acid metabolism, secondary metabolism and stress signal transduction under Cd stress. The Ca application reduced its ABA level but increased the GSH, IAA, ZR and GA3 level, and enhanced its lysine degradation, tyrosine metabolism, alanine, asparagine and glutamate metabolism, plant hormone signal transduction and phenylpropanoid biosynthesis under Cd stress.

Living photosynthetic microneedle patches for in situ oxygenation and postsurgical melanoma therapy.

Surgical excision remains the principal treatment for melanoma, while tumor recurrence and delayed wound healing often occur due to the residual tumor cells and hypoxic microenvironment in the postoperative skin wounds. Herein, we present a living photosynthetic microneedle (MN) patch (namely MA/CM@MN) loaded with microalgae (MA) and cuttlefish melanin (CM) for postsurgical melanoma therapy and skin wound healing. Benefiting from the oxygenic photosynthesis of the alive MA in the MN base, the MA/CM@MN can generate oxygen under light exposure, thus facilitating skin cell proliferation and protecting cells against hypoxia-induced cell death.
