AI Article Synopsis

  • This study investigates the potential of platelet-rich fibrin (PRF) treatment to regenerate damaged endometrium and address intrauterine adhesion (IUA), a leading cause of infertility.
  • Researchers used various methods to analyze PRF's microstructure and established a rat model for IUA, dividing 40 female rats into three groups for comparison.
  • Results showed that PRF facilitated endometrial cell growth, preserved uterine structure, and improved regeneration of the endometrium, suggesting its effectiveness in treating IUA and potential applicability in women's infertility treatments.

Article Abstract

Purpose: Intrauterine adhesion (IUA) is the most common cause of uterine infertility. This study aims to evaluate whether platelet-rich fibrin (PRF) treatment can stimulate damaged endometrium regeneration in rats.

Methods: First, hematoxylin and eosin (HE) staining, scanning and transmission electron microscopy, and ELISAs were used to evaluate the microstructure of PRF. Then, mechanical damage was used to establish an IUA rat model. A total of 40 SD female rats were randomized to three groups: PRF transplantation group, IUA group, and sham group. Rats were sacrificed at 3, 7, and 14 days and uteruses were obtained for further analysis. Finally, functional and histological recovery of the damaged endometrium was analyzed by pregnancy test, HE staining, Masson's staining, and immunohistochemistry.

Results: PRF has two distinct zones, platelets and fibrin zone. Long and narrow fibrin fibers interconnected with each other and formed a three-dimensional, flexible, and elastic structure; platelet aggregates were trapped in fibrin fibers, and each platelet is associated with several fibrin fibers. PRF exudates promoted endometrial stromal cell proliferation and migration . PRF transplantation was beneficial for maintaining uterine structure, promoting endometrial luminal epithelium and endometrial gland regeneration, and decreasing fibrotic areas .

Conclusion: Intrauterine administration of PRF was demonstrated to be effective in preventing IUA and stimulating damaged endometrium regeneration in rats. This study not only provided a promising method for its potential in endometrial regeneration in women who suffer from uterine infertility but also may prevent IUA after intrauterine surgery in clinical cases.

Engraftment of self-renewing endometrial epithelial organoids promotes endometrial regeneration by differentiating into functional glands in rats.

Introduction: Extensive trauma frequently disrupts endometrial regeneration by diminishing endometrial stem cells/progenitor cells, affecting female fertility. While bone marrow mesenchymal stem cell (BMSC) transplantation has been suggested as an approach to address endometrial injury, it comes with certain limitations. Recent advancements in endometrial epithelial organoids (EEOs) have displayed encouraging potential for endometrial regeneration.

Objective: Mitochondria transfer from human Wharton's Jelly-derived mesenchymal stem cells (hWJ-MSCs-mt) and human endometrium-derived mesenchymal stem cells (hE-MSCs-mt), along with curcumin, were explored as potential treatments for age-related macular degeneration (AMD) caused by mitochondrial inefficiency, using a retinal model to assess impacts of curcumin and hWJ-MSCs-mt or hE-MSCs-mt on AMD.

Methods: ARPE-19 cells established an in vitro AMD model. Cells were exposed to 0-50 μM curcumin for 24 hours to determine optimal concentration by assessing their viability.

Unlabelled: Currently, chronic salpingo-oophoritis (CS) is still an urgent problem in the statistics of diseases of the female genital organs. In 35% of women, its course leads to disorder of the fallopian tubes patency and infertility. An additional indirect mechanism of infertility can be the morpho-functional failure of the endometrium.

Zearalenone (ZEA) induces oxidative damage in porcine endometrial stromal cells (ESCs), which is a critical factor affecting the growth and reproduction of female pigs. We hypothesize that rutin, a flavonoid antioxidant, can alleviate ZEA-induced cellular damage through the p53 signaling pathway. In this experiment, porcine ESCs were used as a research model.

Department of Obstetrics and Gynecology, Maruyama Memorial General Hospital, 2-10-5, Honchou, Iwatsuki-ku, Saitama City, Saitama 339-8521, Japan.

Objective: To reduce the damage of uterine endometrium caused during hysteroscopic myomectomy (HM) for reproductive aged patients, a new uterine endometrium preservation hysteroscopic myomectomy (UEP-HM) has been developed. In this study, we introduced this technique with comparing to the conventional hysteroscopic myomectomy (C-HM).

Study Design: The data from 94 patients aged 42 or younger who underwent HM (38 cases with UEP-HM and 56 cases with C-HM) for treating single Type 1 or Type 2 submucosal leiomyoma (SL) were analysed retrospectively for comparing the characteristics of both patient and target SL.
