One-pot multicomponent reaction: An efficient synthesis of 4-(HET)Aryl-substituted pyrazolo-quinolinones.

Introduction: Pyrazolo-quinolinones (PQs) are a versatile class of Organic chemistry and it was found that they have very advanced functionally promising as selective compounds due to their scaffold and demonstrated low toxicity and considerable clinical promise. Moreover, these compounds have an important role in different applications such as analytical reagents, technical, pharmaceutical development, and biological markers. So, we aimed to synthesize and characterize new 4-(Het)aryl-substituted Pyrazole-quinolinones.

Method: Synthesis of One-Pot Multicomponent Reaction was carried out with the reaction of 5-amino-3-methyl-1H-pyrazole 1a, and 5-amino-3-phenyl-1H-pyrazole 1b and dimedone 2 with some (het)aryl aldehydes 3 using ethanol. Characterizations such as the linear structures and the regiospecificity of the reactions were determined by using NMR, GC-MS, IR, and UV measurements.

Conclusion: This research will be very useful for all methods and procedures using multi-one-point reactions which are very popular in the synthesis of heterocyclic systems for pyrazole- quinoline skeleton synthesis.

A straightforward I-promoted Povarov multicomponent reaction strategy for the synthesis of N-heteroaromatic benzothiazoles and 2, 2'-biquinolines was described. This methodology involved a one-pot two-step procedure, tandem iodination/Kornblum oxidation, and [4+2] condensation. This protocol was characterized by broad scope of substrates (49 examples, 31-80 % yields), applicable for large-scale preparation, and downstream derivatization of the products.

This study reports a green, multi-component synthesis of 2-aminoimidazole-linked quinoxaline Schiff bases using a novel superparamagnetic acid catalyst. The catalyst consists of sulfo-anthranilic acid (SAA) immobilized on MnCoFeO@alginate magnetic nanorods (MNRs), achieving high SAA loading (1.8 mmol g) and product yields (91-97%).

This review reports the achievements in copper(II) triflate-catalyzed processes concerning the multicomponent reactions, applied to the synthesis of acyclic and cyclic compounds. In particular, for the heteropolycyclic systems mechanistic insights were outlined as well as cycloaddition and aza-Diels-Alder reactions were included. These strategies have gained attention due to their highly atom- and step-economy, one-step multi-bond forming, mild reaction conditions, low cost and easy handling.

A multi-component method for the synthesis of pyrimido[5,4-]azolo[1,5-]pyrimidines and pyrimido[4,5-][1,2,4]triazolo[5,1-][1,2,4]triazines has been developed. It was shown that vicinal amino-nitrile and amino-ethoxycarbonyl derivatives of azolo[1,5-]pyrimidines and azolo[5,1-][1,2,4]triazines were converted to tricyclic heterocycles in the "AcOH-RC(OEt)-amine" system. Reaction conditions were optimized, patterns of this process were investigated, and intermediates were isolated.
