Analysis of the role of PANoptosis in intervertebral disk degeneration via integrated bioinformatics analysis and experimental validation.

Int Immunopharmacol

December 2024

Department of Orthopedics, The Affiliated Traditional Chinese Medicine Hospital, Southwest Medical University, Luzhou 646000, Sichuan Province, China; Luzhou Longmatan District People's Hospital, Luzhou, Sichuan, China. Electronic address:

Intervertebral disc degeneration (IVDD) is an age-related orthopedic degenerative disease characterized by recurrent episodes of lower back pain, and death of nucleus pulposus cells (NPCs) has been identified as a key factor in the pathophysiological process of IVDD episodes. Recent studies have shown that " PANapoptosis ", a newly characterized form of cell death, has emerged as an important factor contributing to the development of several diseases. However, studies on the specific mechanisms of its role in the development of IVDD are lacking.

Tumor Resection in Hepatic Carcinomas Restores Circulating T Regulatory Cells.

J Clin Med

October 2024

Flow Cytometry Unit, Department of Clinical Pathology, Hospitais da Universidade de Coimbra, Unidade Local de Saúde de Coimbra, 3000-076 Coimbra, Portugal.

Article Synopsis
  • - Cholangiocarcinoma (CCA) and hepatocellular carcinoma (HCC) are significant types of liver cancer that impact the immune response due to the role of T regulatory (Treg) cells in suppressing antitumor immunity.
  • - A study was conducted on healthy individuals, CCA patients, and HCC patients to assess changes in natural killer (NK) cells and T cell subsets before and after surgical tumor removal, using various blood assays.
  • - Findings revealed that both types of cancer patients exhibited alterations in Treg cell populations before surgery, with an increase in certain Treg cells after tumor resection, indicating potential mechanisms of immune modulation following surgery.

Background: Peripheral blood mononuclear cells (PBMCs) serve as the immune system's primary transportation hub outside of the affected ablated tissue. This study aims to explore the transcriptomic profiling of the immune response in PBMCs induced by microwave ablation (MWA) in low-risk thyroid cancer.

Methods: For eight patients diagnosed with low-risk thyroid cancer, 10 ml of peripheral venous blood was collected before MWA as well as one day and one month after MWA.

Overexpression of miR-96 leads to retinal degeneration in mice.

Biochem Biophys Res Commun

July 2024

Eye Hospital, Wenzhou Medical University, Wenzhou 325027, China. Electronic address:

Article Synopsis
  • The combined knockout of miR-183 and miR-96 in mice leads to severe retinal degeneration, while a single knockout of miR-96 only causes developmental delays.
  • Overexpressing miR-96 in the retina, even at safe levels, induces significant degeneration of photoreceptors, evidenced by various analysis techniques.
  • The research indicates that proper levels of miR-96 are crucial for maintaining retinal health and suggests that overexpression triggers immune responses and microglial activation associated with degeneration.

Objectives: This study aimed at revealing the underlying mechanisms of the loss and gain of ceftazidime-avibactam susceptibility in a non-carbapenemase-producing hypervirulent (hvKp).

Methods: Here we longitudinally recovered 3 non-carbapenemase-producing K1-ST23 hvKp strains at a one-month interval (KP29105, KP29499 and KP30086) from an elderly male. Antimicrobial susceptibility testing, whole genome sequencing, transcriptomic sequencing, gene cloning, plasmid conjugation, quantitative real-time PCR (qRT-PCR), and SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis) were conducted.
