α-Synuclein (α-Syn) aggregation into fibrils with prion-like features is intimately associated with Lewy pathology and various synucleinopathies. Emerging studies suggest that α-Syn could form liquid condensates through phase separation. The role of these condensates in aggregation and disease remains elusive and the interplay between α-Syn fibrils and α-Syn condensates remains unexplored, possibly due to difficulties in triggering the formation of α-Syn condensates in cells. To address this gap, we developed an assay allowing the controlled assembly/disassembly of α-Syn condensates in cells and studied them upon exposure to preformed α-Syn fibrillar polymorphs. Fibrils triggered the evolution of liquid α-Syn condensates into solid-like structures displaying growing needle-like extensions and exhibiting pathological amyloid hallmarks. No such changes were elicited on α-Syn that did not undergo phase separation. We, therefore, propose a model where α-Syn within condensates fuels exogenous fibrillar seeds growth, thus speeding up the prion-like propagation of pathogenic aggregates.

An easy-to-synthesize aggregation-induced emission (AIE) active Schiff base HNSA was obtained by condensing equimolar amount of 3-hydroxy-2-naphthohydrazide and salicylaldehyde. In pure DMSO, HNSA is non-fluorescent, but increasing the HEPES (HO, 10 mM, pH 7.4) fraction (f) ≥ 90% showed an intense green fluorescence with maximum fluorescence intensity at 515 nm.

In this paper, we undertake a systematic exploration of soliton turbulent phenomena and the emergence of extreme rogue waves within the framework of the one-dimensional fractional nonlinear Schrödinger (FNLS) equation, which appears in many fields, such as nonlinear optics, Bose-Einstein condensates, plasma physics, etc. By initiating simulations with a plane wave modulated by small noise, we scrutinized the universal regimes of non-stationary turbulence through various statistical indices. Our analysis elucidates a marked increase in the probability of rogue wave occurrences as the system evolves within a certain range of Lévy index α, which can be ascribed to the broadened modulation instability bandwidth.

We demonstrate the application of mechanochemistry in the synthesis of indolone-based photoswitches (hemiindigos, hemithioindigos, and oxindoles) via Knoevenagel condensation reactions. Utilizing ball-milling and an organic base (piperidine) acting as catalyst and solvent for liquid assisted grinding (LAG) conditions, we achieve rapid, solvent-free transformations, obtaining a set of known and previously unreported photoswitches, including highly functional amino acid-based photoswitches, multichromophoric derivatives and photoswitchable cavitands based on resorcin[4]arenes. The reaction under mechanochemical conditions gives moderate-to-high yields and is highly stereoselective leading to Z-isomers of hemiindigos and hemithioindigos and E-isomers of oxindoles.
