Complexity of sample preparation decelerate the development of sample-in-answer-out devices for point-of-need nucleic acid amplification testing. Here, we present the consolidation of alkaline poly(ethylene) glycol-based lysis and solid-phase extraction for rapid and simple sample preparation compatible with direct on-bead amplification. Simultaneous cell lysis and binding of DNA were achieved using an optimised reagent comprising 15% PEG8000, 0.5 M NaCl, and 3.5 mM KOH. This was combined with direct, on-bead amplification using 1.5 μg beads per 20 μL PCR reaction mix. The novel single reagent, 5-min method improved the detection limit by 10 and 100-fold compared with commercial DNA extraction kits and the original alkaline PEG lysis method, respectively. The sensitivity can be further enhanced by one amplification cycle with an ethanol wash or by extending the incubation to 10 min before collecting the magnetic particles. Both methods successfully detected a single copy of Escherichia coli DNA. In biological fluids (saliva, sweat, and urine), the 5-min method was delayed by about one cycle compared to the 15-min method. The proposed methods are attractive for incorporation in the workflow for point-of-need testing of biological samples by providing a practical and chemical method for simple alternative DNA sample preparation.

This study has developed a pressure sensor array based on four functionalized DNA-nanoenzymes with catalase-like activity for multiple detections of foodborne pathogens through a portable pressure manometer. Benefiting from functionalization of 4-mercaptophenylboronic acid and β-mercaptoethylamine, the diversity of nonspecific interactions between four DNA-nanoenzymes and each of the nine bacteria leads to differences in pressure response patterns by catalyzing HO to generate exclusive "fingerprints". As effective statistical tools for processing multivariate data, principal component analysis and hierarchical clustering analysis are employed to identify nine foodborne pathogens by analyzing pressure response patterns.

Developing high-performance alloys is essential for applications in advanced electromagnetic energy conversion devices. In this study, we assess Fe-Co-Ni alloy compositions identified in our previous work through a machine learning (ML) framework, which used both multi-property ML models and multi-objective Bayesian optimization to design compositions with predicted high values of saturation magnetization, Curie temperature, and Vickers hardness. Experimental validation was conducted on two promising compositions synthesized using three different methods: arc melting, ball milling followed by spark plasma sintering (SPS), and chemical synthesis followed by SPS.

This study investigates the properties of egg-free mayonnaise prepared using chia seed protein hydrolysate (CSPH) and pectin extracted from apple pomace (PA) as alternatives to egg, comparing it to traditional egg-based mayonnaise. Chia seed protein was hydrolyzed using Protamex and Bromelain enzymes, while apple pectin was extracted through acid hydrolysis at 90 °C. Four mayonnaise treatments were prepared: T1 (control: 6 % egg), T2 (4 % egg + 1 % CSPH + 1 % PA), T3 (2 % egg + 2 % CSPH + 2 % PA), and T4 (0 % egg + 3 % CSPH + 3 % PA).
