Ascorbate recycling is required for high ascorbate accumulation. Hence, when the ascorbate pool size is small, does the demand for ascorbate recycling decrease? We herein investigate the impact of ascorbate recycling capacity on ascorbate pool size in an ascorbate-deficient background. Our findings demonstrate that a smaller ascorbate pool size lowers the need for ascorbate recycling capacity even under light stress.

The expression profile of the key genes of biosynthesis (VTC2, GPP, GalDH, and GalLDH) and recycling (MDHAR1, MDHAR4, and MDHAR5) of ascorbate in response to infection with the fungal pathogen Fusarium proliferatum in garlic cultivars resistant (Podnebesny) and sensitive (Dubkovsky) to Fusarium rot was determined. It was found that differences in resistance to Fusarium lead to discrepancies in the dynamics and expression of individual genes of the ascorbate pathway, as well as in the ascorbate content. It was shown that, in response to infection, the expression level of the MDHAR4 gene increases in the resistant cultivar and decreases in the Fusarium-sensitive accession.

The human cytochrome b561 (hCytb561) family consists of electron transfer transmembrane proteins characterized by six conserved α-helical transmembrane domains and two β-type heme cofactors. These proteins contribute to the regulation of iron metabolism and numerous different physiological and pathological processes by recycling ascorbic acid and maintaining iron reductase activity. Key members of this family include cytochrome b561 (CYB561), duodenal CYB561 (Dcytb), lysosomal CYB561 (LCytb), stromal cell-derived receptor 2 (SDR2) and 101F6, which are widely expressed in human tissues and participate in the pathogenesis of several diseases and tumors.
