A novel pyrazole migration and cycloaddition process is well developed AgOTf-catalyzed annulation reactions of α-diazo pyrazoleamides with ketimines. This protocol discloses efficient access to synthesize a series of spirooxindole-based β-lactams in good to excellent yields and the diastereoselectivity is switchable by tuning the substituents on the α-diazo pyrazoleamides.

Objective: To investigate the effect of human colorectal fibroblast (CCD-18Co)-conditioned medium (CCD18-Co-CM) on biological behaviors of colorectal cancer (CRC) cells and explore the possible molecular mechanisms.

Methods: Real-time cellular analysis (RTCA), clone formation assay and wound healing assay were used to analyze the changes in proliferation, clone formation, and migration abilities of CRC cell lines HCT116 and Caco-2 treated with CCD18-Co-CM. Western blotting was used to detect the changes in ATK, ERK and STAT3 signaling pathways in the CRC cells activated by CCD18-Co-CM.

Background/aim: Colorectal cancer (CRC) is the third most common malignancy and the second most common cause of cancer-related deaths worldwide. Adavosertib (AZD1775), a small molecule inhibitor of WEE1 kinase, abrogates G/M cell cycle arrest and induces double-stranded DNA breaks. According to previous findings, adavosertib, in combination with other DNA-damaging agents, causes premature mitosis and cell death in p53-mutated cancer cells mainly via abrogation of the G/M cell cycle checkpoint.

Herein, we constructed highly reduced phosphomolybdates crystalline materials - to help this process, with the formula of [Co(CNH)][Co(CNH)(HO)][Co(HPMoO)]·8HO (), [Ni(CNH)(HO)][Ni(CNH)][Ni(HO)][Ni(HPMoO)]·3HO·2CHOH (), and [Zn(CNH)][Zn(CNH)][Zn(CNH)(HO)][Zn(HPMoO)] () [CNH = 2-(1H-pyrazol-3-yl)pyridine]. Specifically, catalyst demonstrated a CO production rate of 3276.

Metal-free radical cascade synthesis of substituted pyrazole derivatives was initiated by PhI(OAc) at 23 °C. This protocol features wide functional group tolerance, a simple purification process without column chromatography, and an oxygen migration. Compound 5 demonstrated significant anti-inflammatory activity, indicating potential for therapy.
