Determining concise sets of genomic markers that identify cell types and states within tissue ecosystems remains challenging. To address this challenge, we developed Recurrent Composite Markers for Biological Identities with Neighborhood Enrichment (RECOMBINE). Validations of RECOMBINE with simulation and transcriptomics data in bulk, single-cell and spatial resolutions demonstrated the method's ability for unbiased selection of composite markers that characterize biological subpopulations. RECOMBINE captured markers of mouse visual cortex from single-cell RNA sequencing data and provided a gene panel for targeted spatial transcriptomics profiling. RECOMBINE identified composite markers of CD8 T cell states including GZMK HAVCR2 effector memory cells associated with anti-PD1 therapy response. The method outperformed differential gene expression analysis in characterizing a rare cell subpopulation within mouse intestine. Using RECOMBINE, we uncovered hierarchical gene programs of inter- and intra-tumoral heterogeneity in breast and skin tumors. In conclusion, RECOMBINE offers a data-driven approach for unbiased selection of composite markers, resulting in improved interpretation, discovery, and validation of cell types and states.

This study investigated the protective effects of the dietary polyphenol vanillic acid (VA) on dextran sulfate sodium-induced acute ulcerative colitis (UC) in mice, focusing on its impact on the gut microbiota and inflammatory responses. VA was supplemented following dextran sulfate sodium administration, and key indicators, including body weight, disease activity index, colon length, spleen index, and inflammatory markers, were assessed. VA supplementation significantly alleviated UC symptoms, preserved intestinal barrier integrity, and reduced pro-inflammatory cytokine levels.

Lymphedema, a severe and complex inflammatory disease caused by lymphatic system insufficiency and impeded lymphatic drainage that causes an enormous physical and psychological burden on patients and may even lead to death, has long been a challenging issue in the medical field. Clinically, conventional approaches including surgical treatment and conservative treatment have been employed for lymphedema therapy, but their curative effect is still unsatisfactory because of high operational difficulty, high cost, and long-term reliance. In this study, a novel kind of piezoelectric microneedle driven by ultrasound (US) is proposed to regulate macrophage polarization and remodel the pathological inflammatory microenvironment in a noninvasive manner, thereby promoting lymphatic regeneration and improving lymphedema.
