The purification and biochemical characterization of the extracellular alpha amylase from MTCC5152 were studied. The combined use of ion exchange and gel filtration chromatographic methods were used for purification studies. The specific activity was significantly increased (33 fold) and 19.41 fold purification of the enzyme -amylase with 24% yield was achieved. The enzyme had an optimal pH of 6.5 and exhibited its highest activity at 55 °C. It is active over a wide range of pH 5-7 at room temperature. The enzyme is relatively stable in the temperature range of 25-35 °C for a period of 4 h hence, more suitable for industrial applications. and value of the enzyme was to be 5.882 mg/mL and 0.803 mg/mL/min respectively using starch as the substrate. The purified protein showed a single band on native and SDS PAGE and the molecular weight was found to be 31 kDa. Starch zymogram also revealed one clear zone of amylolytic activity which corresponded to the band obtained with native PAGE and SDS/PAGE. The characterization studies showed that the enzyme activity is inhibited by Ca, Mn, Hg, Fe.

This study demonstrated a novel approach to accurately estimate 5-day biochemical oxygen demand (BOD) in textile wastewater using a microbial consortium from food processing wastewater fixed on coconut fibers. Although glucose-glutamic acid (GGA) has been widely known as the most preferred substrates for microbial respiration, its calibration surprisingly resulted in an overestimation of BOD in textile wastewater due to its lower utilization rate compared to that of textile wastewater. After being adapted with a new nutrient environment composed of GGA and textile wastewater, the adapted packed-bed bioreactors (PBBRs) was capable of accurate estimation of BOD in textile wastewater using GGA standard solution.

Background: The antigen Na-GST-1, expressed by the hookworm Necator americanus, plays crucial biochemical roles in parasite survival. This study explores the development of mRNA vaccine candidates based on Na-GST-1, building on the success of recombinant Na-GST-1 (rNa-GST-1) protein, currently assessed as a subunit vaccine candidate, which has shown promise in preclinical and clinical studies.
Methodology/findings: By leveraging the flexible design of RNA vaccines and protein intracellular trafficking signal sequences, we developed three variants of Na-GST-1 as native (cytosolic), secretory, and plasma membrane-anchored (PM) antigens.

Zwitterionic polymers have garnered significant attention for their distinctive properties, such as biocompatibility, antifouling capabilities, and resistance to protein adsorption, making them promising candidates for a wide range of applications, including drug delivery, oil production inhibitors, and water purification membranes. This study reports the synthesis and characterization of zwitterionic monomers and polymers through the modification of linear, vinyl, and aromatic heterocyclic functional groups via reaction with 1,3-propanesultone. Four zwitterionic polymers with varying molecular structures-ranging from linear to five and six membered ring systems-were synthesized: poly(sulfobetaine methacrylamide) (pSBMAm), poly(sulfobetaine-1-vinylimidazole) (pSB1VI), poly(sulfobetaine-2-vinylpyridine) (pSB2VP), and poly(sulfobetaine-4-vinylpyridine) (pSB4VP).
