Herein, we provide a protocol for visualizing active osteoclast cathepsin K (CatK) with the quenched-fluorescent-activity-based probe qTJK17. We describe steps for isolating peripheral blood mononuclear cells, their differentiation into osteoclasts, and TRAP staining using an acid phosphatase leukocyte kit. We then detail visualization of active CatK. The probe qTJK17 includes a reactive group, acyloxymethylketone, that binds to the CatK active site, recognition sequence, and fluorescence donor-acceptor pair. This protocol can determine the exact localization of active CatK in osteoclasts. For complete details on the use and execution of this protocol, please refer to Janiszewski et al. (2023)..

Background: Postoperative nausea and vomiting (PONV) after orthognathic surgery remains one of the most common side effects despite the use of several medications.

Purpose: The study aimed to compare the frequencies of PONV between a combination of metoclopramide with granisetron and granisetron alone.

Study Design, Setting, Sample: A randomized double-blind clinical trial was conducted in 66 consecutive patients who underwent orthognathic surgery at the Department of Oral and Maxillofacial Surgery at Bezmialem Vakif University.

Objectives: To investigate the impact of compressed sensing - sensitivity encoding (CS-SENSE) acceleration factor on the diagnostic performance of magnetic resonance imaging (MRI) within standard female pelvis protocol in patients with endometrial cancer.

Methods: T2-weighted turbo spin echo (TSE) sequence from standard female pelvic MRI protocol was chosen due to its long acquisition time and essential role in the evaluation of morphological characteristics of the female pelvic anatomical structures. Fully sampled reference scans and multiple prospectively 2x to 5x under-sampled CS-SENSE scans were acquired.

Appropriate disposal of dead farming animals is required to guarantee effective disease control while protecting the environment. In crisis situations, alternatives to rendering can be used, including on-farm burial. The objectives of this study were to: (i) describe the burial and monitoring protocols used on poultry farms in France in response to major avian influenza outbreaks; (ii) assess the effectiveness of the burial protocol, in terms of both technical and biosecurity aspects, and microbiological, physical and chemical changes of the buried materials and the environment over time; (iii) provide recommendations for future burial and follow-up protocols.

Ex vivo electroretinography (ERG) provides insight into the health and functionality of retinal cells, the integrity of phototransduction, and the visual cycle and allows for the direct application of pharmaceuticals to the retinal tissues. Here, we present a protocol for performing ex vivo ERG on adult zebrafish. We describe steps for zebrafish tissue dissection, mounting tissues, and assembling and connecting cassettes.
