Neoxanthin is undetectable in human blood after ingestion of fresh young spinach leaf.

In a previous study, we demonstrated that the carotenoid neoxanthin was contained in young leafy vegetables including spinach and showed a fat accumulation inhibitory effect in vitro. To evaluate the bioavailability of neoxanthin, a raw young spinach leaf (100 g day-1 for 4 weeks) intake test was performed on 14 participants (36.5 ± 8.0 years; male:female ratio = 9:5). Neoxanthin, neochrome, β-carotene, and lutein concentration in the spinach and blood of participants (before and after the test) was measured using high performance liquid chromatography. Neither neoxanthin nor neochrome was detected in the blood samples, whereas β-carotene and lutein concentration significantly increased (1.4- and 1.9-fold, respectively) during testing. Neoxanthin bioavailability in humans is low; thus, it is unlikely to have a fat accumulation inhibitory effect in vivo, contrary to the result in vitro. Ingesting the leafy vegetables raw can help maintain high neoxanthin levels, but it is not beneficial for neoxanthin bioavailability.

Albino tea cultivars with distinct leaf colors have rarely been used to explore the metabolic mechanism of leaf color formation. Carotenoids and chlorophyll are primary pigments affecting leaf color with differential metabolic mechanisms in albino tea resources. TEM analysis showed that in albino stage, much less chloroplasts with abnormal structure were observed in white and yellow tea leaves, which corresponded to the significant reduction of total chlorophyll, Chl a and Chl b, and increase of Chl a/b, resulting in tea leaves to lose their green color.

Individual carotenoids provide significant health benefits to humans, and potatoes are recognized as some of the most stable crops, distinguished by their substantial carotenoid content. To elucidate the accumulation patterns of individual carotenoids in potatoes, we quantified the carotenoid content in yellow- and white-fleshed genotypes across five developmental stages using LC-MS/MS. A total of 22 carotenoids were identified in yellow potatoes, whereas 18 were detected in white potatoes.

The article reports methods for the expression and assay of 9-cis-epoxycarotenoid cleavage dioxygenase (NCED), an enzyme involved in the biosynthesis of phytohormone abscisic acid in plants. A method for the preparation of the unstable substrate 9'-cis-neoxanthin from fresh spinach is described. The inhibition of Solanum lycopersicum NCED by a series of aryl hydroxamic acid inhibitors is illustrated, and inhibitors D2 and D4 are assayed against NCED isozymes from Zea mays.
