Somatic cell reprogramming and oncogenic transformation share surprisingly similar features, yet transformed cells are resistant to reprogramming. Epigenetic barriers must block transformed cells from reprogramming, but the nature of those barriers is unclear. In this study, we generated a systematic panel of transformed mouse embryonic fibroblasts (MEFs) using oncogenic transgenes and discovered transformed cell lines compatible with reprogramming when transfected with Oct4/Sox2/Klf4/Myc. By comparing the reprogramming-capable and incapable transformed lines we identified multiple stages of failure in the reprogramming process. Some transformed lines failed at an early stage, whilst other lines seemed to progress through a conventional reprogramming process. Finally, we show that MEK inhibition overcomes one critical reprogramming barrier by indirectly suppressing a hyperacetylated active epigenetic state. This study reveals that diverse epigenetic barriers underly resistance to reprogramming of transformed cells.

Purpose: Trophoblast cell-surface antigen 2 (Trop2) is overexpressed in various solid tumors and contributes to tumor progression, while its expression remains low in normal tissues. Trop2-targeting antibody-drug conjugate (ADC), sacituzumab govitecan-hziy (Trodelvy), has shown efficacy in targeting this antigen. We report a green approach to prepare carbon dots (CDs) with fresh tomatoes as carbon sources and amino acids as dopants (ACDs) by a microwave assisted method. The synthesised CDs were analysed by UV-visible absorption spectroscopy, photoluminescence spectroscopy, high resolution transmission electron spectroscopy, X-ray diffraction, Fourier transform infrared spectroscopy, X-ray photo electron spectroscopy. An MTT assay was used to evaluate the cytotoxicity of CDs toward L929 cells and found that CDs exhibit low cytotoxicity.

In Japan, most HbA1c measurements by enzymatic assays or immunoassays represent the HbA1c levels in the blood cell fraction obtained after centrifugation of the blood samples. The present study investigated that the blood cell enzymatic HbA1c assay (EA-HbA1c) was compared with whole blood HbA1c in patients with iron deficiency anemia (IDA). CD7-targeted chimeric antigen receptor-T (CAR-T) cell therapy has shown great promise in the treatment of relapsed/refractory T-cell acute lymphoblastic leukemia (T-ALL). In this study, we reported a case of a 34-year-old male patient with T-ALL who finally developed multi-line drug resistance and refractoriness after multiple lines of high-intensity chemotherapy. After physician evaluation, this patient received allogeneic hematopoietic stem cell transplantation (allo-HSCT).
