This cross-sectional study compared plasma brain-derived neurotrophic factor (BDNF) levels among chronic primary musculoskeletal pain patients, chronic widespread pain patients, and asymptomatic controls. The study included 126 participants aged 18-65, divided into three groups of 42 each. Pain intensity was assessed using a Numeric Rating Scale (NRS), and plasma BDNF levels were measured via ELISA.

Background: Physically inactivity is associated with increased risk of dementia including Alzheimer's disease (AD). Prior work from the Harvard Aging Brain Study (HABS) suggests that lower baseline physical activity in cognitively unimpaired individuals with elevated amyloid burden is associated with faster prospective cognitive decline. However, whether this detrimental effect on cognition in preclinical AD is mediated by accelerated tau pathology remains unclear.

Background: The locus coeruleus (LC)-norepinephrine (NE) system is one of the first systems affected in Alzheimer's Disease (AD), prior to cortical involvement. LC-NE system dysregulation has also been associated with neuropsychiatric and stress-related symptoms, early non-cognitive signals of AD. This study investigates whether structural and functional LC-NE system metrics are associated with affective and stress-related reports among predominantly cognitively healthy adults, and whether these associations are exacerbated by AD fluid biomarkers of tau, neurodegeneration and astrocyte reactivity.

Background: Autophagy-lysosomal pathway (ALP) dysfunction emerges early in Alzheimer's disease(AD). In mouse AD models, lysosomal acidification deficits impair ALP in neurons, in some inducing massive autolysosome accumulations, intraneuronal amyloid plaque, and early neuronal death yielding an extracellular amyloid plaque. This distinctive neurodegenerative pattern (PANTHOS) emerges in early-stage AD and is recapitulated in human late-onset AD (accompanying poster).
