AI Article Synopsis

  • Gold-metallic nanofibrils were created using three iso-apoferritin proteins with varying ratios of light and heavy subunits, ranging from 0% to 100% light subunits.
  • These fibrils can simultaneously nucleate and grow gold nanoparticles, leading to the formation of hybrid inorganic-organic metallic nanowires that follow the helical structure of the protein fibers.
  • The gold nanoparticles within these systems maintained their optical properties, and conductivity tests revealed that the nanowires exhibited ohmic behavior, similar to continuous metallic materials.

Here, we report the X-ray structure of the adduct formed upon reaction of cisplatin, one of the most prescribed anticancer agents for the clinic treatment of solid tumors, with the apo-form of human serum transferrin (hTF). Two Pt binding sites were identified in both molecules of the adduct present in the crystal asymmetric unit: Pt binds close to the side chains of Met256 and Met499 at the N- and C-lobe, respectively. In the crystal structure, the cisplatin moiety bound to Met256 also interacts with Ser616 from a symmetry related molecule.

Different sensitivities of porcine coronary arteries and veins to BAY 60-2770, a soluble guanylate cyclase activator.

Nitric oxide (NO)-donor drugs, which stimulate reduced form of soluble guanylate cyclase (sGC), have different efficacy to the arteries and veins. This study examined whether sGC activators, which activate oxidized/apo sGC, also have arteriovenous selectivity similar to that of NO-donor drugs. The mechanical responses of the isolated blood vessels were assessed using the organ chamber technique and protein expression was verified using western blotting.

Staphylococcus aureus has evolved mechanisms to cope with low iron (Fe) availability in host tissues. Staphylococcus aureus uses the ferric uptake transcriptional regulator (Fur) to sense titers of cytosolic Fe. Upon Fe depletion, apo-Fur relieves transcriptional repression of genes utilized for Fe uptake.

IN SILICO BASED RE-ENGINEERING OF A COMPUTATIONALLY DESIGNED BIOSENSOR WITH ALTERED SIGNALLING MODE AND IMPROVED DYNAMIC RANGE.

A current challenge in the rational design of biomolecular sensors is the ability to custom design binding affinities and detection mode in silico. To this end, we re-engineered a previously reported computationally-designed fluorescent maltooligosaccharide (MOS)-detecting biosensor to both alter its ligand-binding affinity and to analyse the underlying sensing mechanism. The dynamic range of the biosensor was expanded through the computer aided introduction of a series of amino acid substitutions in the starting protein scaffold (MalX from Streptococcus pneumoniae), which generated a biosensor set with binding affinities spanning over five orders of magnitude.
