Complex/cryptic Gene Fusions and 1q Jumping Translocation in Pediatric Ewing Sarcomas. | LitMetric

AI Article Synopsis

Article Abstract

Ewing sarcomas (ES) are rare small round cell sarcomas often affecting children and characterized by gene fusions involving one member of the FET family of genes (usually and a member of the ETS family of transcription factors (usually or ). The detection of rearrangements has important diagnostic value. Here, we conducted a retrospective review of 218 consecutive pediatric ES at diagnosis and found eight patients having data from chromosome analysis, FISH/microarray, and gene-fusion assay. Three of these eight ES had novel complex/cryptic rearrangements/fusions by chromosome analysis. One case had a t(9;11;22)(q22;q24;q12) three-way translocation involving fusion and 1q jumping translocation. Two cases had cryptic rearrangements/fusions, including one case with a cryptic t(4;11;22)(q35;q24;q12) three-way translocation involving fusion, and the other had a cryptic rearrangement/fusion on an abnormal chromosome 22. All patients in this study had various aneuploidies with a gain of chromosome 8 (75%), the most common, followed by a gain of chromosomes 20 (50%) and 4 (37.5%), respectively. Recognition of complex and/or cryptic gene rearrangements/fusions and other chromosome abnormalities (such as jumping translocation and aneuploidies) using a combination of various genetic methods is important for accurate diagnosis, prognosis, and treatment outcomes of pediatric ES.

Article Synopsis
  • Jumping translocations (JT), linked to disease progression in myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML), involve the movement of a tri-tetra-somic 1q chromosome to various other chromosomes.
  • Research showed that in patients with SRSF2 mutations, JT was associated with changes in DNA methylation during treatment with 5'-azacytidine (AZA), revealing significant shifts in the methylome and impacting various biological pathways.
  • The study highlighted that epigenetic modifications, including changes in DNA methylation and specific signaling pathways like PI3K/AKT and MAPK, play a crucial role in the progression of myeloid neoplasms associated with

The systematics of the family Dendrodorididae, with only three valid genera, is a challenge for integrative taxonomists. Its members lack hard structures for morphological comparisons and their mitochondrial and nuclear markers provide contradictory phylogenetic signals, making phylogenetic reconstructions difficult. This molecular discordance has been hypothesized to be the result of nuclear pseudogenes or exogenous contamination.

Jumping translocation (JT) is a cytogenetic event in which a donor chromosomal segment is translocated to two or more recipient chromosomes. We describe a case of a 75-year-old female patient diagnosed with acute myeloid leukemia (AML) with monocytic differentiation having acquired JT involving 3q21→3qter as a donor chromosomal segment with 12 different recipient chromosomes. Each abnormal clone had monosomy 7 and trisomy 8.

Article Synopsis
  • Genetic counseling for mosaicism with balanced translocation at amniocentesis is generally straightforward due to most cases presenting normal phenotypes.
  • In contrast, counseling for mosaicism with unbalanced translocation is complex, as outcomes can vary, resulting in either normal or abnormal phenotypes.
  • The article reviews reported cases of mosaic unbalanced translocation, addressing various genetic factors and challenges for genetic counselors and obstetricians aiding parents considering the outcomes.

1q jumping translocation (JT) is rare and its molecular profiles in myeloid malignancies are not well-known. This study evaluated gene mutations in 1q-JT cohorts (0.38%) from hematological malignant specimens that underwent genetic analysis at the Johns Hopkins Hospital (n = 11,908) and the MD Anderson Cancer Center.
