Article Abstract
The phagosome is a redox-active organelle. Numerous reductive and oxidative systems play both direct and indirect roles in phagosomal function. With the advent of newer methodologies to study these redox events in live cells, the details of how redox conditions change within the maturing phagosome, how they are regulated, and how they influence other phagosomal functions can be investigated. In this chapter, we detail phagosome-specific, fluorescence-based assays that measure disulfide reduction and the production of reactive oxygen species in live phagocytes such as macrophages and dendritic cells, in real time.

As wildfire events become more frequent, there is a need to better understand the impact of smoke on the environment and human health. Smoke, or biomass burning aerosol (BBA), can undergo atmospheric processing changing its chemical and optical properties. We examined the interactions between four lignin pyrolysis products (catechol, syringol, syringic acid, and vanillic acid) and three BBA-relevant iron oxide mineral phases (hematite, maghemite, and magnetite) using attenuated total reflectance-Fourier transform infrared spectroscopy and dissolved iron measurements to better understand how atmospheric processing changes concentrations of soluble iron, iron oxidation state, and brown carbon abundance.

In this study, ex-situ catalytic pyrolysis of oxygen-containing polycarbonate (PC) was conducted to prepare carbon nanotubes (CNTs) and H-rich syngas. This study examined the influence of the active metal components (Ni and Fe), catalyst pre-reduction, and pre-deoxygenation of pyrolysis volatiles on the catalytic performance and mechanism. Results show that the reductive constituents in pyrolysis volatiles make it difficult to reduce the Fe oxides, thus hindering the CNTs growth on Fe catalysts, compared to Ni catalysts.

Anthracycline doxorubicin (DOX) remains the first-line chemotherapeutic drug for the efficient treatment of breast cancer, but its severe cardiotoxicity limits its long-term application in clinical tumor chemotherapy. Until now, the pathogenesis mechanism of DOX-induced cardiotoxicity (DIC) is still not fully understood. Glutathione (GSH) is an abundant thiol-containing tripeptide that functions in redox homeostasis, protein folding, and iron metabolism. In Saccharomyces cerevisiae, GSH depletion leads to increased sensitivity to oxidants and other toxic compounds, disruption of Fe-S cluster biogenesis, and eventually cell death. GSH pools are supplied by intracellular biosynthesis and GSH import from the extracellular environment.

A dual photoredox/cobalt-catalyzed protocol for chemo-, regio-, diastereo- and enantioselective reductive coupling of 1,1-disubstituted allenes and cyclobutenes through chemo-, regio-, diastereo- and enantioselective oxidative cyclization followed by stereoselective protonation promoted by a chiral phosphine-cobalt complex is presented. Such process represents an unprecedented reaction pathway for cobalt catalysis that enables selective transformation of the less sterically congested alkenes of 1,1-disubstituted allenes with cyclobutenes, incorporating a broad scope of tetrasubstituted alkenes into the cyclobutane scaffolds in up to 86% yield, >98:2 chemo- and regioselectivity, >98:2 dr and >99.5:0.
