Role of synthetic coordination chemistry in pharmaceutical science is expeditiously increased due to its sundry relevances in this field. The present review endows the synthesized macrocyclic complexes of transition metal ions containing isatin and its derivatives as ligand precursors, their characterization and their copious pharmaceutical applications. Isatin (1-Indole-2,3-dione) is a protean compound (presence of lactam and keto moiety permits to change its molecular framework) that can be obtained from marine animals, plants, and is also found in mammalian tissues and in human fluids as a metabolite of amino acids. It can be used for the synthesis of miscellaneous organic and inorganic complexes and for designing of drugs since it has remarkable utility in pharmaceutical industry due to its wide range of biological and pharmacological activities, for instance anti-microbial, anti-HIV, anti-tubercular, anti-cancer, anti-viral, anti-oxidant, anti-inflammatory, anti-angiogenic, analgesic activity, anti-Parkinson's disease, anti-convulsant etc. This review provides extensive information about the latest methods for the synthesis of isatin or its substituted derivatives based macrocyclic complexes of transition metals and their plentiful applications in medicinal chemistry.

The reaction between 1,3-bis(3,5-dimethylpyrazolylmethyl)hexahydropyrimidine L and Mo(CO) in CHCN at 130 °C afforded a binuclear Mo(0) complex 1 containing a new macrocycle formed upon C-N bond cleavage in L in good yield. Conversely, a clean reaction takes place between L and [Mo(CO)(COD)] in THF at 60 °C to give a new metalloligand complex [Mo(CO)(κ-,-L)] 2 containing a spectator pyrazole arm in 83% yield. Their structures were determined by X-ray diffraction methods, and a plausible mechanism is proposed for the C-N bond cleavage leading to complex 1.

Compartmentalized models with coupled catalytic networks are considered as "protocells" in the context of research related to the origin of life. To model the kinetics of a simple cellular uptake-metabolism process, we use a compartmentalized protocell system that combines liposome-encapsulated intravesicular reporter pairs with co-encapsulated enzymes to monitor the membrane transport of a substrate (analyte uptake) and its subsequent enzymatic reaction inside the vesicles (metabolism to the product). The intravesicular chemosensing ensembles consist of the macrocycles cucurbit[7]uril or p-sulfonatocalix[4]arene and matching fluorescent dyes to set up suitable reporter pairs.

Designing the architecture of donor-acceptor (D-A) pairs is an effective strategy to tailor the electronic structure of conjugated macrocycles for optoelectronic devices. Herein, we present the synthesis of three D-A nanohoops ( = 7, 8, 9) containing a naphthalene diimide (NDI) unit as an acceptor and []cycloparaphenylenes ([]CPPs) moieties as donors. The D-A characteristics of were substantiated through absorption and fluorescence spectroscopic studies, electrochemical investigations, and computational analysis.

Air-stable single-molecule magnets (SMMs) can be obtained by confining Dy ion in a coordination environment; however, most of the current efforts were focused on modifying the rigidity of the macrocycle ligand. Herein, we attempt to assemble air-stable SMMs based on macrocycles with a replaceable coordination site. By using an in situ 1 + 1 Schiff-base reaction of dialdehyde with diamine, three air-stable SMMs have been obtained in which one of the equatorial coordination sites can be varied from -NH- (for ), -O- (for ), and -NMe- (for ).

A bromophenylpyridine derivative (N1) was designed, synthesized, and the molecule was incorporated into the cavity of the cucurbit[8]uril (Q[8]) as a guest to form a 2:1 host-guest complex. This complex demonstrates good room temperature phosphorescence (RTP) properties in aqueous solution. The host-guest interaction and optical properties of N1@Q[8] in aqueous solution were studied by means of H NMR, ultraviolet-visible absorption spectroscopy, fluorescence spectroscopy, phosphorescence spectroscopy, scanning electron microscopy and inverted fluorescence microscopy.
