The therapeutic effects of human mesenchymal stromal cells (MSC) have been attributed mostly to their paracrine activity, exerted through small-secreted extracellular vesicles (EVs) rather than their engraftment into injured tissues. Currently, the production of MSC-derived EVs (MSC-EVs) is performed in laborious static culture systems with limited manufacturing capacity using serum-containing media. In this work, a serum-/xenogeneic-free microcarrier-based culture system was successfully established for bone marrow-derived MSC cultivation and MSC-EV production using a 2 l-scale controlled stirred tank reactor (STR) operated under fed-batch (FB) or fed-batch combined with continuous perfusion (FB/CP). Overall, maximal cell numbers of (3.0 ± 0.12) × 10 and (5.3 ± 0.32) × 10 were attained at Days 8 and 12 for FB and FB/CP cultures, respectively, and MSC(M) expanded under both conditions retained their immunophenotype. MSC-EVs were identified in the conditioned medium collected from all STR cultures by transmission electron microscopy, and EV protein markers were successfully identified by Western blot analysis. Overall, no significant differences were observed between EVs isolated from MSC expanded in STR operated under the two feeding approaches. EV mean sizes of 163 ± 5.27 nm and 162 ± 4.44 nm (p > 0.05) and concentrations of (2.4 ± 0.35) × 10 EVs/mL and (3.0 ± 0.48) × 10 EVs/mL (p > 0.05) were estimated by nanoparticle tracking analysis for FB and FB/CP cultures, respectively. The STR-based platform optimized herein represents a major contribution toward the development of human MSC- and MSC-EV-based products as promising therapeutic agents for Regenerative Medicine settings.

As Streptomycetes might produce melanin to survive in stressful environmental conditions, like under metal exposure, supplementing metal ions to the growth medium could be a wise strategy for boosting the production of the pigment. The aim of this study was to test, for the first time, the possibility of boosting DSM40314 melanin biosynthesis by adding to the growth medium singularly or, at the same time, different concentrations (1.0, 1.

This work investigated the production and characterization of a silk fibroin (SF) hydrogel incorporated with an (AV) extract. Four extraction methods, ultrasound-assisted extraction with bath and probe, stirring, and Soxhlet, were tested, while the hydrogel was produced by a one-step freeze-thaw method. Besides the extraction yield, the antioxidant capacity of the extracts was accessed, which allowed to select the extract obtained by ultrasound-assisted extraction to be incorporated into the hydrogels.

Porous CuNi films are promising candidates for electrocatalytic water splitting, with their catalytic performance largely influenced by the crystallographic structure and chemical state. In this study, by employing a magnetic field-controlled bubble template-assisted electrodeposition method, CuNi films with a preferred Ni(111) crystal orientation were synthesized. Moreover, adjusting the magnetic field direction during deposition can affect the degree of preferred orientation and, consequently, the electrochemical activity of the films.

This study investigates the formation and properties of vesicles produced via biocatalytic Polymerization-Induced Self-Assembly (bioPISA) as artificial cells. Methods for achieving size uniformity, including gentle centrifugation and sucrose gradient centrifugation, are explored, and the effects of stirring speed on vesicle morphology is investigated. The internal structure of the vesicles, characterized by a polymer-rich matrix, is analyzed using fluorescence correlation spectroscopy (FCS).

Stimulus-responsive liposomes (L) are increasingly recognized for their potential in enhancing therapies, especially in cancer nanomedicine, owing to their ability to encapsulate drugs of diverse properties efficiently. In this study, a quality-by-design (QbD) strategy was proposed to optimize the surface functionalization of gold nanoparticles (AuNPs) on doxorubicin (Dox)-loaded L intended for improving cancer treatment. Thin-film hydration and pH-gradient methods were applied for L preparation and Dox loading, respectively.
