In addition to the action potentials used for axonal signaling, many neurons generate dendritic "spikes" associated with synaptic plasticity. However, in order to control both plasticity and signaling, synaptic inputs must be able to differentially modulate the firing of these two spike types. Here, we investigate this issue in the electrosensory lobe (ELL) of weakly electric mormyrid fish, where separate control over axonal and dendritic spikes is essential for the transmission of learned predictive signals from inhibitory interneurons to the output stage of the circuit. Through a combination of experimental and modeling studies, we uncover a novel mechanism by which sensory input selectively modulates the rate of dendritic spiking by adjusting the amplitude of backpropagating axonal action potentials. Interestingly, this mechanism does not require spatially segregated synaptic inputs or dendritic compartmentalization but relies instead on an electrotonically distant spike initiation site in the axon-a common biophysical feature of neurons.

Unlabelled: Human umbilical cord-derived mesenchymal stromal cells (UC-MSCs), which can be prepared in advance and are presumed to be advantageous for nerve regeneration, have potential as a cell source for Bio 3D conduits. The purpose of this study was to evaluate the nerve regeneration ability of Bio 3D conduits made from UC-MSCs using a rat sciatic nerve defect model.
Methods: A Bio 3D conduit was fabricated using a Bio 3D printer by placing UC-MSC spheroids into thin needles according to predesigned 3D data.
Unlabelled: The corticospinal tract (CST) facilitates skilled, precise movements, which necessitates that subcerebral projection neurons (SCPN) establish segmentally specific connectivity with brainstem and spinal circuits. Developmental molecular delineation enables prospective identification of corticospinal neurons (CSN) projecting to thoraco-lumbar spinal segments; however, it remains unclear whether other SCPN subpopulations in developing sensorimotor cortex can be prospectively identified in this manner. Such molecular tools could enable investigations of SCPN circuitry with precision and specificity.

The frequent poor functional outcomes after delayed surgical repair of injured human peripheral nerves results in progressive downregulation of growth-associated genes in parallel with reduced neuronal regenerative capacity under each of the experimental conditions of chronic axotomy of neurones that remain without target contact, chronic distal nerve stump denervation, and chronic muscle denervation. Brief (1 h) low-frequency (20 Hz) electrical stimulation (ES) accelerates the outgrowth of regenerating axons across the surgical site of microsurgical repair of a transected nerve. Exercise programmes also promote nerve regeneration with the combination of ES and exercise being the most effective.
