Objectives: Pyroptosis-related genes (PRGs) are abnormally expressed in a variety of gastrointestinal tumors, this study aimed to investigate the role of pyroptosis genes in assessing the prognosis of esophageal cancer (ESCA).
Methods: Through consensus clustering, we identified two subtypes associated with PRGs. After Lasso regression and multivariate Cox regression analysis, a polygenic signature based on six prognostic PRGS was constructed. Afterwards, we combined the risk score with clinical predictors to construct and validate a PRGs-associated ESCA prognostic model.
Results: Through analysis, we Successfully constructed and validated a PRGs-associated ESCA prognostic model that predicts ESCA survival and correlates with the tumor immune microenvironment.
Conclusion: Based on PRGs features, we established a new ESCA hierarchical model. This model has important clinical implications for ESCA patients, both in terms of assessing prognosis and in terms of targeted and immunotherapy.

Pyroptosis and macrophage pro-inflammatory activation play an important role in hepatocellular carcinoma (HCC) progression. However, the specific regulatory mechanisms remain unclear. We identified pyroptosis-related differentially expressed genes (DEGs) based on the GSE4183 and GSE44861 datasets as well as EVenn database.

HERVs (Human endogenous retroviruses) are remnants of ancient exogenous retroviruses that have integrated into the human genome, particularly in germ-line cells. Among these, the envelope protein gene (Human endogenous retroviruses W family envelope protein), located on chromosome 7 and primarily expressed in the human placenta, has been closely linked to various neuropsychiatric disorders, including schizophrenia, as well as autoimmune diseases and cancer. Recent studies have highlighted the abnormal expression of cytokines as a key factor in the pathophysiology of schizophrenia.

Lung cancer is a highly prevalent and fatal cancer that seriously threatens the safety of people in various regions around the world. Difficulty in early diagnosis and strong drug resistance have always been difficulties in the treatment of lung cancer, so the prognosis of lung cancer has always been the focus of scientific researchers. This study used genotype-tissue expression (GTEx) and the cancer genome atlas (TCGA) databases to obtain 477 lung adenocarcinoma (LUAD) and 347 healthy individuals' samples as research subjects and divided LUAD patients into low-risk and high-risk groups based on prognostic risk scores.

Osteosarcoma is a malignant bone tumor originating from mesenchymal tissue. Recent studies have found that the tumor inflammatory microenvironment plays an important role in promoting the malignant characteristics and metastatic potential of malignant tumors. Pyroptosis, an inflammatory programmed cell death, elicits immune responses that exhibit anti-tumor effects through released factors and contents.

Background: The role of pyroptosis in lung squamous cell carcinoma (LUSC) remains unclear. This study aimed to screen pyroptosis-related genes (PRGs) and construct a model to investigate the immune infiltration, gene mutations, and immune response of patients of LUSC.
Methods: We conducted a comprehensive evaluation of pyroptosis patterns in patients with LUSC with 51 PRGs.
