This work reports some observations on the fine structure of the different types of Paracentrotus lividus permanent blastulae obtained after treatment with BUdR. Some permanent blastulae appeared similar to normal early mesenchyme blastulae and at the ultrastructural level they did not show any particular differences compared with the normal embryos. The only remarkable fact was that nucleoli often displayed the fibrous portion still abundant. Other permanent blastulae showed the blastocoel partially or completely, invaded by cells and it was often very difficult to recognize the embryo polarity. In these embryos, at the ultrastructural level, the most striking effects were observed in the blastocoelic cells. On the basis of the results reported in this paper and of the data already existing in the literature about the effects of BUdR on sea urchin embryos, the possibility is discussed that the drug might affect selectively some steps essential for the realizing of the embryo differentiation.

Early interspecies nuclear transfer (iNT) experiments suggested that a foreign nucleus may become permanently damaged after a few rounds of cell division in the cytoplasm of another species. That is, in some distant species combinations, nucleocytoplasmic hybrid (cybrid) blastula nuclei can no longer support development, even if they are back-transferred into their own kind of egg cytoplasm. We monitored foreign DNA amplification and RNA production by quantitative PCR (qPCR) and RT-qPCR in interorder amphibian hybrids and cybrids formed by the transfer of newt (Pleurodeles waltl) embryonic nuclei into intact and enucleated frog (Xenopus laevis) eggs.

We examined several candidate posterior/mesodermal inducing molecules using permanent blastula-type embryos (PBEs) as an assay system. Candidate molecules were injected individually or in combination with the organizer factor chordin mRNA. Injection of chordin alone resulted in a white hemispherical neural tissue surrounded by a large circular cement gland, together with anterior neural gene expression and thus the development of the anterior-most parts of the embryo, without mesodermal tissues.

The tandemly repeated sea urchin alpha-histone genes are developmentally regulated. These genes are transcribed up to the early blastula stage and permanently silenced as the embryos approach gastrulation. As previously described, expression of the alpha-H2A gene depends on the binding of the MBF-1 activator to the 5' enhancer, while down-regulation relies on the functional interaction between the 3' sns 5 insulator and the GA repeats located upstream of the enhancer.

Ablation of vegetal cytoplasm from newly fertilized Xenopus eggs results in the development of permanent blastula-type embryos (PBEs). PBEs cleave normally and develop into a very simple tissue consisting only of atypical epidermis.
