The development of single-cell multiomics has provided the ability to systematically investigate cellular diversity and heterogeneity in different biological systems via comprehensive delineations of individual cellular states. Single-cell RNA sequencing in particular has served as a powerful tool to the study of the molecular circuitries underlying preimplantation embryonic development in both the mouse and human. Here we describe a method to elucidate the cellular dynamics of the embryo further by performing both single-cell RNA sequencing (Smart-Seq2) and single-cell small non-coding RNA sequencing (Small-Seq) on the same individual embryonic cell.

Diabetes nephropathy (DN) is a prevalent and severe microvascular diabetic complication. Despite the recent developments in germacrone-based therapies for DN, the underlying mechanisms of germacrone in DN remain poorly understood. This study used comprehensive bioinformatics analysis to identify critical microRNAs (miRNAs) and the potential underlying pathways related to germacrone activities.

The inclusion of microalgae in functional fish diets has a notable impact on the welfare, metabolism and physiology of the organism. The microbial communities associated with the fish are directly influenced by the host's diet, and further understanding the impact on mucosal microbiota is needed. This study aimed to analyze the microbiota associated with the skin and gills of Sparus aurata fed a diet containing 10% microalgae.

Recently, RNA velocity has driven a paradigmatic change in single-cell RNA sequencing (scRNA-seq) studies, allowing the reconstruction and prediction of directed trajectories in cell differentiation and state transitions. Most existing methods of dynamic modeling use ordinary differential equations (ODE) for individual genes without applying multivariate approaches. However, this modeling strategy inadequately captures the intrinsically stochastic nature of transcriptional dynamics governed by a cell-specific latent time across multiple genes, potentially leading to erroneous results.

Variation of gene ratios in mock communities constructed with purified 16S rRNA during processing.

Sci Rep

16S ribosomal nucleic acid (16S rRNA) analysis allows to specifically target the metabolically active members of microbial communities. The stability of the ratios between target genes in the workflow, which is essential for the bioprocess-relevance of the data derived from this analysis, was investigated using synthetic mock communities constructed by mixing purified 16S rRNA from Bacillus subtilis (Bs), Staphylococcus aureus (Sa), Pseudomonas aeruginosa (Pa), Klebsiella pneumoniae (Kp) and Burkholderia cepacia (Bc) in different proportions. The RT reaction yielded one copy of cDNA per rRNA molecule for Pa, Bc and Sa but only 2/3 of the expected cDNA from 16S rRNAs of Bs and Kp.
