TnpB is a member of the Obligate Mobile Element Guided Activity (OMEGA) RNA-guided nuclease family, is harbored in transposons, and likely functions to maintain the transposon in genomes. Previously, it was shown that TnpB cleaves double- and single-stranded DNA substrates in an RNA-guided manner, but the biogenesis of the TnpB ribonucleoprotein (RNP) complex is unknown. Using purified apo TnpB, we demonstrate the ability of TnpB to generate guide omegaRNA (ωRNA) from its own mRNA through 5' processing. We also uncover a potential -regulatory mechanism whereby a region of the TnpB mRNA inhibits DNA cleavage by the RNP complex. We further expand the characterization of TnpB by examining ωRNA processing and RNA-guided nuclease activity in 59 orthologs spanning the natural diversity of the TnpB family. This work reveals a new functionality, ωRNA biogenesis, of TnpB, and characterizes additional members of this biotechnologically useful family of programmable enzymes.

The unique fuel characteristics of butanol and the possibility of its microbial production make it one of the most desirable environmentally friendly substitutes for petroleum fuels. However, the highly toxic nature of 1-butanol to the bacterial strains makes it unprofitable for commercial production. By comparison, 2-butanol has similar fuel qualities, and despite the difficulties in its microbial synthesis, it holds promise because it may be less toxic.

Article Synopsis
  • - Cas12 proteins are diverse nucleic acid-targeting proteins that utilize RNA guides and a single nuclease domain to target DNA and occasionally RNA.
  • - Their high variability is the result of different evolutionary paths from transposon-related ancestors, leading to significant differences in size, structure, and function among various Cas12 variants.
  • - The article reviews the unique characteristics of both natural and engineered Cas12 proteins, outlines their applications in genome editing and diagnostics, and explores future research directions.

Collateral nuclease activity of TnpB triggered by high temperature enables fast and sensitive nucleic acid detection.

Commun Biol

November 2024

State Key Laboratory of Biocatalysis and Enzyme Engineering, Hubei Key Laboratory of Industrial Biotechnology, School of Life Sciences, Hubei University, Wuhan, PR China.

Article Synopsis
  • TnpB proteins from the IS200/IS605 family have potential for genome editing, but their uncharacterized collateral nuclease activity hinders the development of diagnostic tools.
  • Research revealed that TnpB from a heat-loving archaeon has increased ability to cleave single-stranded DNA when heated, and mutations in target DNA affect this activity.
  • A new detection method called TESD (TnpB Enable fast and Sensitive Detection) was created, achieving sensitivity of 29 copies per microliter in 30 minutes, paving the way for improved molecular diagnostics.
