Swellable Core Technology (SCT) tablets, a solid oral dosage formulation designed for the controlled release of Active Pharmaceutical Ingredient (API), are made up of two distinct layers; an active layer containing the active ingredient (10-30%wt) and up to 90%wt polyethylene oxide (PEO); and a sweller layer which contains up to 65%wt PEO. The objective of this study was to develop a process to remove PEO from analytical test solutions and optimize API recovery using physicochemical properties of the API. Quantitation of PEO was performed by liquid chromatography (LC) using an evaporative light scattering detector (ELSD). This was used to build an understanding of removal of PEO using solid-phase extraction and liquid-liquid extraction techniques. A workflow was proposed to allow efficient development of analytical methods for SCT tablets with optimized sample clean-up.

Cadmium (Cd) removal from soil is crucial as Cd enters the food chain and affect food safety, thus impose severe threaten to human health. We developed PPC@PC-Fe, a dual-functional core-shell sphere for efficient soil Cd reduction. The shell (PPC) was composed of encapsulated citric acid (CA) in a polylactic acid (PLA) and polyethylene glycol (PEG) network, which endows a function of activating Cd; and the core (PC-Fe) consisted of a polyacrylic acid/carboxymethyl chitosan (PAA/CMC) hydrogel with FeO nanoparticles to adsorb adjacent activated Cd.

Colon Drug Delivery Systems Based on Swellable and Microbially Degradable High-Methoxyl Pectin: Coating Process and In Vitro Performance.

Pharmaceutics

April 2024

Department of Pharmaceutical Sciences, Section of Pharmaceutical Technology and Legislation "M.E. Sangalli", University of Milan, Via G. Colombo 71, 20133 Milan, Italy.

Oral colon delivery systems based on a dual targeting strategy, harnessing time- and microbiota-dependent release mechanisms, were designed in the form of a drug-containing core, a swellable/biodegradable polysaccharide inner layer and a gastroresistant outer film. High-methoxyl pectin was employed as the functional coating polymer and was applied by spray-coating or powder-layering. Stratification of pectin powder required the use of low-viscosity hydroxypropyl methylcellulose in water solution as the binder.

IDO-triggered swellable polymeric micelles for IDO inhibition and targeted cancer immunotherapy.

Indoleamine 2,3-dioxygenase (IDO) has been studied as a promising target for cancer immunotherapy. IDO catalyzes the oxidation of tryptophan into kynurenine, which subsequently activates regulatory T cells, thereby promoting an immunosuppressive microenvironment in the tumor tissue. Due to its overexpression in tumor cells, IDO itself could be a tumor-specific stimulus for targeted cancer therapy.

In the present study, timed-release indomethacin tablets, releasing drug after predetermined lag times, were developed for the effective treatment of early morning stiffness in rheumatoid arthritis using two-nozzle fused deposition modeling (FDM) 3D printing with a Bowden extruder. The developed core-shell tablets consisted of a drug-containing core and release-regulating shell with different designed thicknesses (i.e.
