AI Article Synopsis

  • Vitamin K (K1 and K2) and Vitamin D are essential for bone health, particularly in relation to osteoporosis, and measuring their levels in the blood can help understand their roles in skeletal muscle metabolism.
  • A new, efficient high-performance liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed to precisely measure serum levels of VK1, MK-4, MK-7, 25(OH)D2, and 25(OH)D3 in a cohort of 200 patients.
  • Results showed that VK2 (MK-7) levels decreased earlier than vitamin D in postmenopausal women with osteoporosis, suggesting that MK-7 could be a useful biomarker

Article Abstract

Introduction: Vitamin K (VK) as well as vitamin D (VD) plays an important role in osteoporosis. Vitamin K1 (VK1), vitamin K2 (VK2, menaquinone-4 (MK-4), and menaquinone-7 (MK-7)) are significant for the metabolism of skeletal muscle. 25-hydroxyvitamin D2 and 25-hydroxyvitamin D3 (25(OH)D2 and 25(OH)D3) reflect circulating VD levels. More sensitive measurements remain to be developed. In the present study, a new high-performance liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for the determination of VK1, VK2 (MK-4 and MK-7), as well as 25(OH)D2 and 25(OH)D3 levels in human serum.

Methods: We developed a simple LC-MS/MS method for the determination of VK1, MK-4, MK-7, 25(OH)D2, and 25(OH)D3 levels in human serum and validated the method in a study cohort of 200 patients divided into the premenopausal women group and postmenopausal osteoporosis patient group.

Results: The overall precision (coefficient of variation) ranged from 2.66 to 10.11% in the specified working range (0.05-5 ng/mL) for VK1, MK-4, and MK-7. Serum VK1, MK-4, and MK-7 levels in postmenopausal women with osteoporosis were 1.187 ± 0.094 ng/mL, 0.058 ± 0.009 ng/mL, and 0.885 ± 0.064 ng/mL, respectively (mean ± standard deviation). Serum VK1, MK-4, and MK-7 levels in premenopausal women were 1.143 ± 0.103 ng/mL, 0.028 ± 0.003 ng/mL, and 1.553 ± 0.226 ng/mL, respectively. Serum 25(OH)D2 and 25(OH)D3 levels in postmenopausal women with osteoporosis were 0.757 ± 0.056 ng/mL and 11.72 ± 0.632 ng/mL, respectively. Serum 25(OH)D2 and 25(OH)D3 levels in premenopausal were 1.793 ± 0.575 ng/mL and 12.42 ± 1.069 ng/mL, respectively.

Conclusion: A new LC-MS/MS method for determination of serum VK and VD levels was evaluated and validated. MK-7 in plasma decreased earlier than VD in postmenopausal osteoporosis patients. MK-7 status is significantly associated with osteoporosis and could be considered a predictable biomarker in the diagnosis of osteoporosis in postmenopausal women.

Study on Vitamin K Levels in Mature Milk of Chinese Lactating Mothers.

Nutrients

October 2024

Key Laboratory of Public Nutrition and Health, National Health Commission of the People's Republic of China, Beijing 100050, China.

Objectives: This study sought to understand the levels of vitamin K in the mature milk of Chinese lactating mothers, thereby providing a foundation for the development of appropriate intake (AI) of vitamin K for infants aged 0-5 months.

Methods: Five hundred healthy lactating mothers were selected from the mature milk bank established by the Chinese Maternal and Infant Nutrition and Health Cohort by using a simple random sample procedure. Relevant information about lactating mothers and their infants was obtained by a questionnaire survey.

Article Synopsis
  • There are differences in bioactivity among various forms of vitamin K (VK), which impact anticoagulation therapy.
  • A study established a method to measure levels of VK1 and two forms of VK2 (MK-4 and MK-7) in plasma samples from cardiovascular patients.
  • Results showed that concentrations of MK-7 and total VKs were significantly associated with initial anticoagulation levels (INR0), suggesting these vitamin K forms should be considered when adjusting anticoagulation doses.

Short-term impact of vitamin K2 supplementation on biochemical parameters and lipoprotein fractions.

Electrophoresis

August 2024

Department of Medical Chemistry and Clinical Biochemistry, Second Faculty of Medicine, Charles University in Prague, Motol University Hospital, Prague, Czechia.

This study explored the short-term effects of vitamin K2 (VK2) supplementation on biochemical parameters (vitamin D, vitamin E, vitamin A, alkaline phosphatase, calcium, phosphorus (P), magnesium, metallothionein, triglycerides, cholesterol, high-density lipoprotein (HDL), low-density lipoprotein (LDL), and lipoprotein fractions (albumin, HDL, very low-density lipoprotein (VLDL), LDL, and chylomicrons). A short-term experiment (24 h, six probands) was performed to track changes in VK2 levels after a single-dose intake (360 µg/day). Liquid chromatography-tandem mass spectrometry was used to monitor vitamin K levels (menaquinone-4 (MK-4), menaquinone-7 (MK-7), and vitamin K1 [VK1]) with a limit of detection of 1.
