Benzylisoquinoline alkaloids in lotus () seed plumules and leaves exhibit significant tissue specificity for their pharmacological effects and potential nutritional properties. Herein, 46 benzylisoquinoline alkaloids were identified via UPLC-QTOF-HRMS, of which 9 were annotated as glycosylated monobenzylisoquinoline alkaloids concentrated in the seed plumules. The spatial distribution of targeted benzylisoquinoline alkaloids in leaves, seed plumules, and milky sap was determined via MALDI-MSI. Furthermore, 37 cultivars were investigated using targeted metabolomics to provide insights into functional tea development. While aporphine alkaloids comprised the main compounds present in lotus leaves, bisbenzylisoquinoline alkaloids were the main compounds in lotus plumules, where glycosylation primarily occurs. These findings can help understand the distribution of benzylisoquinoline alkaloids in lotus tissue and the directional breeding of varieties enriched with specific chemical functional groups for nutritional and pharmacological applications.

How well is the female population represented in clinical trials with infusion therapies for Parkinson's disease? A systematic review and metanalysis.

Eur J Neurol

January 2025

Parkinson and Movement Disorders Unit, Study Center for Neurodegenerative diseases (CESNE), Department of Neuroscience, Padua Neuroscience Center (PNC), University of Padua, Padua, Italy.

Background: Parkinson's disease (PD) is a neurodegenerative disorder affecting both sexes, but differences exist between male and female in clinical manifestations, functional impact of symptoms and hormonal influences. Therefore, representativeness of females in PD trials indirectly determines the external validity of the clinical research in this field.

Objective: To estimate the representativeness of female in infusion therapy trials for advanced PD.

Benzylisoquinoline alkaloids are significant phyto-nutraceuticals with a bitter flavor. However, due to their complex structures, the sensing and identification of benzylisoquinoline alkaloids is challenging. Benzylisoquinoline alkaloids are commonly extracted from plants such as Coptidis Rhizoma (CR), while the adulteration and dyeing of CR are prevalent.

Introduction: In Poland, not all forms of device-aided therapies for advanced Parkinson's Disease (APD) are currently available.

Material And Methods: We aimed to produce a consensus recommendation from Polish movement disorders experts after discussing gaps in the APD care pathway in Poland.

Results: Rescue therapy with apomorphine (APO) PEN injection and levodopa-entacapone-carbidopa intestinal gel infusion are not included in Poland's Specialist Therapeutic Programme, and are thus not reimbursed.

Berberine enhances the antibacterial activity of thymoquinone, carvacrol and thymol against multi-drug resistant nontuberculous mycobacteria.

Pol J Vet Sci

September 2024

Department of Biology and Fish Diseases, Faculty of Veterinary Medicine, University of Life Sciences in Lublin, Akademicka 12, 20-033 Lublin, Poland.

The aim of this study was to investigate the activity of thymoquinone (TQ), carvacrol (CAR) and thymol (TYM) against multi-drug resistant nontuberculous mycobacteria (MDR-NTM), alone and in combination with berberine (BER). Antimicrobial activity was first evaluated at concentrations from 8 to 512 μg/mL. Each of the compounds tested exhibited good activity against nontuberculous mycobacteria (NTM) isolated from fish, with MIC values of 32-128 μg/mL.

Berberine alleviates AGEs-induced ferroptosis by activating NRF2 in the skin of diabetic mice.

Exp Biol Med (Maywood)

December 2024

Institute of Disease-Oriented Nutritional Research, Guangzhou Red Cross Hospital, Jinan University, Guangzhou, China.

Advanced glycation end products (AGEs) have adverse effects on the development of diabetic complications. Berberine (BBR), a natural alkaloid, has demonstrated its ability to promote the delayed healing of skin wounds. However, the impact of BBR on AGEs-induced ferroptosis in skin cells and the underlying molecular mechanisms remains unexplored.
