Hydrogen sulfide (HS) plays a cytoprotective role during mitophagy by detoxifying superfluous reactive oxygen species (ROS), and its concentration fluctuates in this process. However, no work has been reported to reveal the variation in HS levels during autophagic fusion of lysosomes and mitochondria. Herein, we present a lysosome-targeted fluorogenic probe, named NA-HS, for real-time monitoring of HS fluctuation for the first time. The newly synthesized probe exhibits good selectivity and high sensitivity (detection limit of 23.6 nM). Fluorescence imaging results demonstrated that NA-HS could image exogenous and endogenous HS in living cells. Interestingly, the colocalization results revealed that the level of HS was upregulated after autophagy began because of the cytoprotective effect, and was finally gradually reduced during subsequent autophagic fusion. This work not only affords a powerful fluorescence tool to monitor the variations in HS levels during mitophagy, but also offers new insights into targeting small molecules for elaborating the complex cellular signal pathways.

The aim of this study was to investigate the inhibitory effect of nintedanib (BIBF) on glioblastoma (GBM) cells and its mechanism of action and to optimize a drug delivery strategy to overcome the limitations posed by the blood-brain barrier (BBB). We analyzed the inhibition of GBM cell lines following BIBF treatment and explored its effect on the autophagy pathway. The cytotoxicity of BIBF was assessed using the CCK-8 assay, and further techniques such as transmission electron microscopy, Western blotting (WB), and flow cytometry were employed to demonstrate that BIBF could block the autophagic pathway by inhibiting the fusion of autophagosomes and lysosomes, ultimately limiting the proliferation of GBM cells.

Here, we questioned the role of presenilins (PSENs 1 and 2), previously shown to be involved in autophagy regulation, in the secretion of small endocytic-originating extracellular vesicles known as exosomes. Indeed, while wild-type cells responded to stimuli promoting both multivesicular endosome (MVE) formation and secretion of small extracellular vesicles (sEVs) enriched in canonical exosomal proteins, PSEN-deficient cells were almost unaffected to these stimuli.

Bisphenol A (BPA) is a common endocrine disruptor chemical that is widely used in the production of food plastic packaging, and it has been shown to potentially harm the reproductive system. However, the specific mechanism by which BPA induces apoptosis of Leydig cells (LCs) and inhibits testosterone synthesis in these cells is unclear. In the present study, TM3 cells were used as an experimental model in combination with a reactive oxygen species (ROS) scavenger (N-acetylcysteine), Caspase-3 inhibitor (Ac-DEVD-CHO), autophagy activator (Torin2), and autophagy inhibitor (Chloroquine) to investigate the potential mechanisms by which BPA causes TM3 cell damage in vitro.
