CRISPR-Cas12a based one-pot detection system has been used in nucleic acid detection and diagnosis. However, it is not sensitive enough to distinguish single nucleotide polymorphisms (SNP), which has greatly restricted its application. To overcome these limitations, we engineered a LbCas12a variant with enhanced sensitivity against SNP, named seCas12a (sensitive Cas12a). SeCas12a-based one-pot SNP detection system is a versatile platform that could use both canonical and non-canonical PAM, and was almost not limited by mutation types to distinguish SNPs located between position 1 to 17. The use of truncated crRNA further improved SNP specificity of seCas12a. Mechanistically, we found only when the -cleavage was at low level between 0.01min and 0.0006 min, a good signal-to-noise ratio can be achieved in one-pot test. SeCas12a-based one-pot SNP detection system was applied to detect pharmacogenomic SNPs in human clinical samples. Of thirteen donors tested in two different SNPs, the seCas12a mediated one-pot system could faithfully detect the SNPs in 30 min with 100% accuracy.

Single nucleotide polymorphism (SNP), as one of the key components of the genetic factors, is important for disease detection and early screening of hereditary diseases. Current SNP genotyping methods require laboratory instruments or long operating times. To facilitate the diagnosis of hereditary diseases, we developed a new method referred to as the LwaCas13a-based SNP genotyping platform (Cas13a platform), which is useful for detecting disease-related SNPs.

Covalent surface modification of silica nanoparticles (SNPs) offers great potential for the development of multimodal nanomaterials for biomedical applications. Herein, we report the synthesis of covalently conjugated bifunctional SNPs and their application to multimodal imaging. Bis(methallyl)silane 15 with cyclopropene and maleimide, designed as a stable bifunctional linker, was efficiently synthesized by traceless Staudiger ligation, and subsequently introduced onto the surface of monodispersed SNPs Sc(OTf)-catalyzed siloxane formation.

Amine modified nano-silica was prepared via a one-pot route and under very mild conditions in water in oil microemulsion with a non-ionic surfactant to study the effect of changing the amount of N-[3-(Trimethoxysilyl)propyl]ethylenediamine (DA) added to the synthesis mixture on the characteristics of the obtained nanocomposite such as morphology, crystallinity, surface charge, particle size, surface area, and accordingly the effect of all of these factors on the efficiency of the nanocomposite for the removal of heavy metal ions, namely zinc, from aqueous solutions. XRD, SEM, TGA, BET, DLS, FTIR, and pH analysis were performed for samples and the results showed a strong effect for the amount of DA added to the synthesis mixture on the characteristics of the obtained nanocomposites. It was found that increasing the amount of DA added to the synthesis mixture increased the pH, hydrodynamic particle size obtained by dynamic light scattering analysis, and the particle size obtained by SEM.
