An electrochemiluminescence immunosensor with double co-reaction accelerators based on AgPO@EuPO-AgNP for detecting squamous cell carcinoma antigen.

This study aimed to design a sandwich electrochemiluminescence (ECL) immunosensor with double co-reaction accelerators for sensitively detecting squamous cell carcinoma antigen (SCCA). First, silver orthophosphate (AgPO) nanoparticles were modified on the surface of EuPO nanowires to improve their poor dispersibility/solubility. At the same time, EuPO was used as a co-reaction accelerator to catalyze SO to produce more intermediates (SO), significantly enhancing the ECL signal of AgPO. Ag nanoparticles (AgNP) modified on AgPO@EuPO composite nanomaterials were used not only as linkers of luminescence groups and biomarkers but also as a co-reaction accelerator to effectively enhance ECL signal. The designed ECL immunosensor displayed several advantages, including good stability and reproducibility. Under the optimal conditions, its linear range in detecting SCCA was 0.0001-50 ng·mL, the detection limit was 25 fg·mL (S/N = 3), the recovery was 96.6-100.4%, and the relative standard deviation was less than 4.8%. It was successfully applied to detect SCCA in human serum.

A novel label-free electrochemical immunosensor for the ultrasensitive determination of cancer antigen (CA724) was developed using a novel composite of CoAl layered double hydroxides (CoAl-LDH) and carboxyl-functionalized multiwall carbon nanotubes (MWCNTs-COOH) as platform and ferrocenecarboxylic acid (Fc-COOH) as signal label. The MWCNTs-COOH/Fc-COOH@CoAl-LDH composite was prepared by a convenient and simple one-step ultrasonic method, and various characterization techniques consisting of scanning electron microscopy (SEM), transimission electronic microscopy (TEM), TEM-Mapping, fourier transform infrared (FT-IR), X-ray diffraction (XRD) and X-ray photoelectronic energy spectrum (XPS) were applied to study the size and morphological features. Due to its large specific surface area and multilayer structure, the CoAl-LDH can be post-doped to embed a large amount of signal probe to realize the amplification of the internal reference signal Fc.

An in-situ nanozyme signal tag combined with a DNA-mediated universal antibody-oriented strategy was proposed to establish a high-performance immunosensing platform for Alzheimer's disease (AD)-related biomarker detection. Briefly, a Zr-based metal-organic framework (MOF) with peroxidase (POD)-like activity was synthesized to encapsulating the electroactive molecule methylene blue (MB), and subsequently modified with a layer of gold nanoparticles on its surface. Herein, we have manufactured a newly designed bifunctional impedimetric and amperometric immunosensor for rapidly detecting erythromycin (ERY) in complicated environments and food stuffs. For this, bimetallic cobalt/cerium-layered double hydroxide nanosheets (CoCe-LDH NSs), which was derived from Co-based zeolite imidazole framework via the structure conversion, was simultaneously utilized as the bioplatform for anchoring the ERY-targeted antibody and for modifying the gold and screen printed electrode. Basic characterizations revealed that CoCe-LDH NSs was composed of mixed metal valences, enrich redox, and abundant oxygen vacancies, facilitating the adhesion on the electrode, the antibody adsorption, and the electron transfers.
