Programmable and modular systems capable of orthogonal genomic and transcriptomic perturbations are crucial for biological research and treating human genetic diseases. Here, we present the minimal versatile genetic perturbation technology (mvGPT), a flexible toolkit designed for simultaneous and orthogonal gene editing, activation, and repression in human cells. The mvGPT combines an engineered compact prime editor (PE), a fusion activator MS2-p65-HSF1 (MPH), and a drive-and-process multiplex array that produces RNAs tailored to different types of genetic perturbation.

Objective: Depression is a potential health killer. As an important means of preventing various human diseases, physical exercise plays an important role in reducing the risk of depression. Using data from the Chinese Household Tracking Survey, this study analyzed the mechanisms by which physical exercise, self-rated health and life satisfaction reduce the risk of depression.

Despite the widespread exploration and availability of parcellations for the functional connectome, parcellations designed for the structural connectome are comparatively limited. Current research suggests that there may be no single "correct" parcellation and that the human brain is intrinsically a multiresolution entity. In this work, we propose the Continuous Structural Connectivitity-based, Nested (CoCoNest) family of parcellations-a fully data-driven, multiresolution family of parcellations derived from structural connectome data.
