Fifty percent of the overall operational expenses of biorefineries are incurred during enzymatic-saccharification processes. Cellulases have a global-market value of $1621 USD. Dearth of conventional lignocelluloses have led to the exploration of their waste stream-based, unconventional sources. Native fungus-employing cellulase-production batches fail to yield sustained enzyme titers. It could be attributed to variations in the enzyme-production broth's quasi-dilatant behavior, its fluid and flow properties; heat and oxygen transfer regimes; kinetics of fungal growth; and nutrient utilization. The current investigation presents one of the first-time usages of a substrate mixture, majorly comprising disposed COVID-19 personal protective-equipment (PPE). To devise a sustainable and scalable cellulase-production process, various variable-regulated, continuous-culture auxostats were performed. The glucose concentration-maintaining auxostat recorded consistent endoglucanase titers throughout its feeding-cum-harvest cycles; furthermore, it enhanced oxygen transfer, heat transfer co-efficient, and mass transfer co-efficient by 91.5, 36, and 77%, respectively. Substrate-characterization revealed that an unintended, autoclave-based organsolv pretreatment caused unanticipated increases in endoglucanase titers. The cumulative lab-scale cellulase-production cost was found to be $16.3. The proposed approach is economical, and it offers a pollution-free waste management process, thereby generating carbon credits.

Endophytic fungi, as plant symbionts, produce an elaborate array of enzymes for efficient disintegration of lignocellulosic biomass into constituent monomeric sugars, making them novel source of lignocellulolytic CAZymes with immense potential in future biorefineries. The present study reports lignocellulolytic enzymes production potential of an endophytic halotolerant Penicillium oxalicum strain isolated from Citrus limon, under submerged and solid-state fermentation (SmF & SSF, respectively), in the presence and absence of salt (1 M NaCl). The comparative QTOF-LC/MS-based exoproteome analysis of the culture extracts unveiled differential expression of CAZymes, with the higher abundance of GH6 and GH7 family cellobiohydrolase in the presence of 1 M salt.

Biomass pretreatment for the production of second-generation (2G) ethanol and biochemical products is a challenging process. The present study investigated the synergistic efficiency of purified carboxymethyl cellulase (CMCase), β-glucosidase, and xylanase from Aspergillus fumigatus JCM 10253 in the hydrolysis of alkaline-pretreated sugarcane bagasse (SCB). Lignocellulose was directly used in itaconic acid production by a model filamentous fungus Neurospora crassa. The promoters of two clock control genes and cellobiohydrolase 1 gene were selected for heterologous genes expression by evaluating different types of promoters. The effect of overexpression of different cellulase was compared, and it was found that expression of cellobiohydrolase 2 from Trichoderma reesei increased the filter paper activity by 2 times, the cellobiohydrolase activity by 4.

The present study reports a highly thermostable β-glucosidase (GH3) from Rasamsonia emersonii that was heterologously expressed in Pichia pastoris. Extracellular β-glucosidase was purified to homogeneity using single step affinity chromatography with molecular weight of ~ 110 kDa. Intriguingly, the purified enzyme displayed high tolerance to inhibitors mainly acetic acid, formic acid, ferulic acid, vanillin and 5-hydroxymethyl furfural at concentrations exceeding those present in acid steam pretreated rice straw slurry used for hydrolysis and subsequent fermentation in 2G ethanol plants.
