Monkeypox (MPOX) is a zoonotic viral disease caused by the Monkeypox virus (MPXV), which has become the most significant public health threat within the genus since the eradication of the Variola virus (VARV). Despite the extensive attention MPXV has garnered, little is known about its clinical manifestations in humans. In this study, a high-throughput RNA sequencing (RNA-seq) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) approach was employed to investigate the transcriptional and metabolic responses of HEK293T cells to the MPXV A5L protein.

Article Synopsis
  • The monkeypox outbreak declared a public health emergency in 2022 escalated in 2024 due to a new strain identified in the Democratic Republic of the Congo and surrounding countries.
  • The review utilized databases like PubMed and WHO to analyze the prevalence, treatment, and prevention strategies related to mpox, highlighting its rising cases and fatalities in African nations.
  • The new mpox strain, primarily spreading through sexual networks, poses a significant global health threat, emphasizing the need for a coordinated international response to control the outbreak effectively.

The three members of the genus capripoxvirus (CaPV), lumpy skin disease virus (LSDV), sheeppox virus (SPPV), and goatpox virus (GTPV) have common hosts and areas of overlapping geographical distribution with Rift Valley fever virus (RVFV). Hence, to ensure more cost-effective disease surveillance we developed and evaluated a Luminex assay for the simultaneous detection of antibodies against CaPV and RVFV in domestic ruminants. In cattle, the assay had a sensitivity (Se) of 98.

Article Synopsis
  • Monkeypox (MPOX) is caused by the Orthopoxvirus monkeypox (MPXV) and presents symptoms akin to smallpox, with ongoing outbreaks raising global public health concerns, especially in the Democratic Republic of the Congo and recently in China.
  • A new lineage of MPXV (C.1.1) was identified from samples in a Nantong outbreak, possibly linked to cases imported from Japan.
  • Genomic analysis revealed reduced mutations in MPXV and specific mutations in proteins that could affect its evolution and vaccine development, indicating potential research directions for understanding the virus.

Detecting Monkeypox Virus by Immunohistochemistry.

J Cutan Pathol

December 2024

Department of Dermatology, University of Virginia, Charlottesville, Virginia, USA.

Background: Mpox (formerly known as monkeypox), a zoonotic disease caused by Monkeypox virus (MPXV), has become an international outbreak since May 2022. Mpox often presents with a mild systemic illness and a characteristic vesiculopustular skin eruption. In addition to molecular testing, histopathology of cutaneous lesions usually shows distinctive findings, such as epidermal necrosis, balloon degeneration, papillary dermal edema, and focal dermal necrosis, which have proven helpful in the diagnosis of mpox.
