Disulfide bond rearrangement is a common occurrence during protein analysis or treatment. A convenient and rapid method has been developed to investigate heat-induced disulfide rearrangement of lactoglobulin using matrix-assisted laser desorption/ionization-in-source decay (MALDI-ISD) technology. By analyzing heated lactoglobulin in reflectron and linear mode, we demonstrated that cysteines C66 and C160 exist as free residues other than linked ones in some protein isomers. This method provides a straightforward and expeditious way to assess the cysteine status and structural changes of proteins under heat stress.

Self-healing optically transparent polyimides have potential applications in optoelectronic device fabrication. In this study, for the first time, we successfully prepared a novel self-healing polyimide film containing reversible disulfide bonds through chemical imidization by introducing cystamine as a self-healing functional monomer into the molecular structure of conventional polyimides. The incorporation of cystamine enabled the films to maintain high transmittance (>87%) and tensile strength (>99 MPa).

The increasing demand for high-performance strain sensors has driven the development of innovative composite systems. This study focused on enhancing the performance of composites by integrating liquid metal, carbon nanotubes, and polydimethylsiloxane (PDMS) in an innovative approach that involved advanced interface engineering, filler synergy, and in situ cross-linking of PDMS in solution. Surface modification of liquid metal with allyl disulfide and hydrogen-containing polydimethylsiloxane significantly improved its stability and dispersion within the polymer matrix.

In the food industry, the emulsifying process alters both the stability and quality of the emulsified products prepared by bovine bone high-temperature hydrolysate (BBHH). The microstructure and interactions of BBHH emulsion were characterized by cryo-scanning electron microscopy (Cryo-SEM) and Raman spectroscopy during emulsification. Notably, BBHH emulsion exhibited the best properties under emulsifying for 120 s, attributed to its interfacial adsorption characteristics.

Protein translocation is a highly dynamic process and, in addition, mitochondrial protein import is especially complicated as the majority of nuclear encoded precursor proteins must engage with multiple translocases before they are assembled in the correct mitochondrial subcompartment. In this chapter, we describe assays for engineered disulfide bond formation and cysteine specific crosslinking to analyze the rearrangement of translocase subunits or to probe protein-protein interactions between precursor proteins and translocase subunits. Such assays were used to characterize the translocase of the outer membrane, the presequence translocase of the inner membrane and the sorting and assembly machinery for the biogenesis of β-Barrel proteins.
