The arylation of sterically hindered amines represents one of the long-standing challenges in synthetic chemistry. Herein, we report a highly efficient Ni-catalysed arylation of sterically hindered primary and secondary amines with aryl chlorides or phenol derivatives enabled by an unsymmetric N-heterocyclic carbene (NHC) ligand. The protocol provides general, efficient, and scalable access to various sterically demanding anilines in excellent yields under mild conditions. A wide range of functional groups and heterocycles are compatible (>50 examples), including those present in biologically relevant molecules. Computational studies suggest that the unsymmetric bulky and flexible NHC ligand was critical to balance the oxidative addition and reductive elimination elementary steps, thus promoting this challenging transformation.

A series of Au(I) complexes containing unsymmetrical N-heterocyclic carbene (imidazolylidene and benzimidazolylidene) functionalized with a xyloside group and an alkyl moiety (methyl and mesityl) was prepared using efficient procedures from D-xylose. Their characterization was carried out in solution by multinuclear NMR, HR-MS spectrometry and cyclic voltammetry, as well as in the solid state by means of single crystal X-ray diffraction analysis for two of them. Evaluation of their ability to inhibit bacterial growth showed a preference for a Gram-positive strain, Staphylococcus aureus, over a Gram-negative strain, Pseudomonas aeruginosa.

The deployment of metalloclusters in applications such as catalysis and materials synthesis requires robust methods for site-differentiation: the conversion of clusters with symmetric ligand spheres to those with unsymmetrical ligand spheres. A simplified and stepwise synthetic method for producing sterically congested unsymmetrical 1,2-dicarbonyl radicals was successfully demonstrated including detailed characterization of each radical cation. Using this approach, an aryl- and N-heterocyclic carbene-substituted 1,2-dicarbonyl radical in its neutral form is generated, revealing the stabilizing role of N-heterocyclic carbenes.
