The global SARS-CoV-2 pandemic caused significant social and economic disruption worldwide, despite highly effective vaccines being developed at an unprecedented speed. Because the first licensed vaccines target only single B-cell antigens, antigenic drift could lead to loss of efficacy against emerging SARS-CoV-2 variants. Improving B-cell vaccines by including multiple T-cell epitopes could solve this problem. Here, we show that predicted MHC class I/II ligands induce robust T-cell responses and protect against severe disease in genetically modified K18-hACE2/BL6 mice susceptible to SARS-CoV-2 infection.

A protein subunit vaccine comprising conserved surface-exposed outer membrane proteins (SE-OMPs) is considered a promising platform for leptospirosis vaccine. The search for novel vaccine candidates that confer high protective efficacy against leptospirosis is ongoing. The LIP3228 protein was previously identified as a conserved and abundant SE-OMP with the potential to serve as an effective vaccine candidate.

Unlabelled: Urinary tract infections (UTIs), largely caused by uropathogenic (UPEC), are increasingly resistant to antibiotics and frequently recur. Using immunoinformatics, we designed a multiepitope peptide vaccine targeting UPEC virulence factors, including iron acquisition systems and adhesins. The construct features 12 cytotoxic T lymphocyte epitopes, six helper T lymphocyte epitopes, and six B-cell epitopes,and isoptimized for high antigenicity, immunogenicity, nontoxic, and low allergenic potential.
