Natural moisturizing factor (NMF) includes several compounds in the stratum corneum (SC), among them, urocanic acid (UCA). Ultraviolet (UV) exposure turns the -UCA of the SC into its isomer. We investigated the impact of a topical emollient emulsion treatment on the UCA isomers of the SC exposed to artificial UV stress. Aliquots of emollient emulsion were applied in healthy subjects for 2 h on delimited areas of the volar forearm, then, the SC was removed by tape stripping. Tapes were irradiated in a solar simulator chamber and a high performance liquid chromatograph was used to quantify UCA isomers from stripped SC extract. The amount of both UCA isomers were almost twice higher in the SC treated with the emollient emulsion. We also observed that the UV irradiation elevated the amount of the / UCA ratio on the SC (non-treated and treated), suggesting that the emollient sample was not able to avoid the UCA isomerization. The in vivo tests corroborated with the UCA data obtained ex vivo, since we found an increase in the superficial skin hydration with respective reduction of the TEWL, probably occurring by the occlusion performed by the emollient emulsion containing 15.0% / of caprylic/capric triglyceride.

L. extract is a promising natural agent for the treatment of atopic dermatitis. It significantly reduces inflammation due to its immunomodulatory properties, mainly attributed to the presence of pentacyclic triterpenes, namely madecassoside and asiaticoside.

Article Synopsis
  • - The study tested the long-term effectiveness and safety of a topical emollient (V0034CR) for treating uremic xerosis and chronic kidney disease-related itching, comparing it to a vehicle control in a phase 3 trial.
  • - Over 28 days, patients applying V0034CR showed a significant improvement in xerosis severity and a greater reduction in scaling compared to those using the control, with 60.2% of the V0034CR group responding positively.
  • - While xerosis symptoms returned without treatment after a break, V0034CR helped maintain remission over a longer period, with high patient satisfaction and minimal side effects like minor irritation.

Chemical warfare agents, particularly vesicants like lewisite, pose a threat due to their ability to cause skin damage through accidental exposure or deliberate attacks. Lewisite rapidly penetrates the skin, causing inflammation and blistering. This study focuses on developing a cream formulation of a therapeutic agent, called integrated stress response inhibitor (ISRIB), to treat lewisite-induced injuries.

Alternatives to Conventional Topical Dosage Forms for Targeted Skin Penetration of Diclofenac Sodium.

Int J Mol Sci

Research & Innovation, Seppic, 127 Chemin de la Poudrerie, 81100 Castres, France.

Skin penetration of an active pharmaceutical ingredient is key to developing topical drugs. This penetration can be adjusted for greater efficacy and/or safety through the selection of dosage form. Two emerging dosage forms, cream-gel and gel-in-oil emulsion, were tested for their ability to deliver diclofenac into the skin, with the target of maximising skin retention while limiting systemic exposure.
