The aim of this study was to develop an innovative, dual-stimuli-responsive smart hydrogel local drug delivery system (LDDS), potentially useful as an injectable simultaneous chemotherapy and magnetic hyperthermia (MHT) antitumor treatment device. The hydrogels were based on a biocompatible and biodegradable poly(-caprolactone---lactide)--poly(ethylene glycol)--poly(-caprolactone---lactide) (PCLA-PEG-PCLA, PCLA) triblock copolymer, synthesized via ring-opening polymerization (ROP) in the presence of a zirconium(IV) acetylacetonate (Zr(acac)) catalyst. The PCLA copolymers were successfully synthesized and characterized using NMR and GPC techniques. Furthermore, the gel-forming and rheological properties of the resulting hydrogels were thoroughly investigated, and the optimal synthesis conditions were determined. The coprecipitation method was applied to create magnetic iron oxide nanoparticles (MIONs) with a low diameter and a narrow size distribution. The magnetic properties of the MIONs were close to superparamagnetic upon TEM, DLS, and VSM analysis. The particle suspension placed in an alternating magnetic field (AMF) of the appropriate parameters showed a rapid increase in temperature to the values desired for hyperthermia. The MIONs/hydrogel matrices were evaluated for paclitaxel (PTX) release in vitro. The release was prolonged and well controlled, displaying close to zero-order kinetics; the drug release mechanism was found to be anomalous. Furthermore, it was found that the simulated hyperthermia conditions had no effect on the release kinetics. As a result, the synthesized smart hydrogels were discovered to be a promising antitumor LDDS, allowing simultaneous chemotherapy and hyperthermia treatment.

Intraocular lenses (IOLs) play a pivotal role in restoring vision following cataract surgery. The evolution of polymeric biomaterials has been central to addressing challenges such as biocompatibility, optical clarity, mechanical stability, and resistance to opacification. This review explores essential requirements for IOL biomaterials, emphasizing their ability to mitigate complications like posterior capsule opacification (PCO) and dysphotopsias while maintaining long-term durability and visual quality.

An imbalance in the body's pH or temperature may modify the immune response and result in ailments such as autoimmune disorders, infectious diseases, cancer, or diabetes. Dual pH- and thermo-responsive carriers are being evaluated as advanced drug delivery microdevices designed to release pharmaceuticals in response to external or internal stimuli. A novel drug delivery system formulated as hydrogel was developed by combining a pH-sensitive polymer (the "biosensor") with a thermosensitive polymer (the delivery component).

Many tissues exhibit structural anisotropy, which imparts orientation-specific properties and functions. However, recapitulating the cellular patterns found in anisotropic tissues presents a remarkable challenge, particularly when using soft and wet hydrogels. Herein, we develop self-assembled anisotropic magnetic FeO micropatterns on polyethylene glycol hydrogels utilizing dipole-dipole interactions.

Polyelectrolyte hydrogels are smart materials whose swelling behavior is governed by ionizable groups on their polymeric chains, making them sensitive to pH and ionic strength. This study combined experiments and modeling to characterize anionic hydrogels. Mechanical tests and gravimetric analyses were performed to track hydrogel mass over time and at a steady state under varying pH and salt concentrations.

Considering the complexity in terms of design that characterizes the different tissues of the human body, it is necessary to study and develop more precise therapies. In this sense, this article presents the possibility of fabricating photocurable thermosensitive hydrogels with free geometry and based on N-Vinyl Caprolactam (VCL) with the aim of modulating the adhesion of non-planar cell cultures. The fabrication process is based on the use as a mold of two-layer thick water-soluble polyvinyl alcohol (PVA) previously printed by Extrusion Material (MatEx).
