Epileptic networks are characterized by two states, seizures or more prolonged interictal periods. Here, we present the procedure for labeling seizure-activated and interictal-activated neuronal ensembles in mouse hippocampal kindling model using an enhanced-synaptic-activity-responsive element. We describe the seizure model establishment, tamoxifen induction, electrical stimulation, and calcium signal recording of labeled ensembles. This protocol has demonstrated dissociated calcium activities in the two ensembles during focal seizure dynamics and can be applied to other animal models of epilepsy. For complete details on the use and execution of this protocol, please refer to Lai et al. (2022)..

α-Synuclein (Syn) is an intrinsically disordered protein, abundant in presynaptic neurons. It is a constituent of the Lewis Body inclusions as amyloid fibrils, in Parkinson's disease patients. It populates an ensemble of conformations and floats between the free random coil and the membrane-bound α-helical species.

We investigate the aging transition in networks of excitable and self-oscillatory units as the fraction of inherently excitable units increases. Two network topologies are considered: a scale-free network with weighted pairwise interactions and a two-dimensional simplicial complex with weighted scale-free pairwise and triadic interactions. Without triadic interactions, the aging transition from collective oscillations to oscillation death (inhomogeneous stationary states) can occur either suddenly or through an intermediate state of partial oscillation.

According to the World Health Organization, the number of people suffering from depressive disorders worldwide is approaching 350 million. The consequences of depressive disorders include considerable worsening of the quality of life, which frequently leads to social isolation. One of the key factors which may cause depression in adulthood is early life stress, in particular, insufficient maternal care during infancy.

Integrate-and-fire models are an important class of phenomenological neuronal models that are frequently used in computational studies of single neural activity, population activity, and recurrent neural networks. If these models are used to understand and interpret electrophysiological data, it is important to reliably estimate the values of the model's parameters. However, there are no standard methods for the parameter estimation of Integrate-and-fire models.

Circuit-based biomarkers distinguishing the gradual progression of Lewy pathology across synucleinopathies remain unknown. Here, we show that seeding of α-synuclein preformed fibrils in mouse dorsal striatum and motor cortex leads to distinct prodromal-phase cortical dysfunction across months. Our findings reveal that while both seeding sites had increased cortical pathology and hyperexcitability, distinct differences in electrophysiological and cellular ensemble patterns were crucial in distinguishing pathology spread between the two seeding sites.
