Calmodulin (CaM) is known to function as a central signal transducer in calcium-mediated intracellular pathways. In this study, a fusion molecule of a recently developed proximity biotinylation enzyme (AirID) with rat CaM (AirID-CaM) was expressed and purified to near homogeneity using an E. coli expression system to examine the physical interactions between CaM and its target proteins by converting the interaction to biotinylation of CaM targets under nondenatured conditions. AirID-CaM catalyzed a Ca-dependent biotinylation of a target protein kinase (Ca/CaM-dependent protein kinase kinase α/1, CaMKKα/1) in vitro, which was suppressed by the addition of excess amounts of CaM, and AirID alone did not catalyze the biotinylation of CaMKKα/1, indicating that the biotinylation of CaMKKα/1 by AirID-CaM likely occurs in an interaction-dependent manner. Furthermore, we also observed the Ca-dependent biotinylation of GST-CaMKIα and GST-CaMKIV by AirID-CaM, suggesting that AirID-CaM can be useful for the rapid detection of CaM/target interactions with relatively high sensitivity.

Time-resolved fluorescence immunochromatographic test strips (TRFIS) was developed for the rapid detection of hepatocellular carcinoma (HCC)-specific plasma exosomes (hExos) by targeting the hExo-surface membrane protein glypican-3 (GPC3). The GPC3-TRFIS could directly detect plasma exosomes without the isolation and purification process, and the whole immunoassay could be completed within 15 min. The visual detection limit of GPC3-TRFIS was 3.

Objectives: To compare the value of musculoskeletal ultrasound (MSUS) with conventional radiography in the detection of patients with early rheumatoid arthritis (RA) and to correlate the sonographic findings with disease activity, and functional disability scores.
Methods: Patients >18 years of age with RA ≤2 years who satisfied the 2010 EULAR/ACR classification criteria for rheumatoid arthritis and disease activity score 28 (DAS28) >2.6, were enrolled.

Digital PCR (dPCR) has transformed nucleic acid diagnostics by enabling the absolute quantification of rare mutations and target sequences. However, traditional dPCR detection methods, such as those involving flow cytometry and fluorescence imaging, may face challenges due to high costs, complexity, limited accuracy, and slow processing speeds. In this study, SAM-dPCR is introduced, a training-free open-source bioanalysis paradigm that offers swift and precise absolute quantification of biological samples.

Proton (H) NMR spectroscopy presents a powerful tool for biomass mixture studies by revealing the involved chemical compounds with identified ingredients and molecular structures. However, conventional H NMR generally suffers from spectral congestion when measuring biomass mixtures, particularly biomass carbohydrate samples, that contain various physically and chemically similar compounds. In this study, a targeted detection NMR approach, DREAMTIME, is exploited for studying biomass carbohydrate mixtures by spectroscopically targeting the desired compounds in separate 1D NMR spectra.
