T-regulatory-type-1 (TR1) cells are a subset of interleukin-10-producing but Foxp3 Treg cells that arise in response to chronic antigenic stimulation. We have shown that systemic delivery of autoimmune disease-relevant peptide-major histocompatibility complex class II (pMHCII)-coated nanoparticles (pMHCII-NP) triggers the formation of large pools of disease-suppressing Foxp3 TR1 cells from cognate T-follicular helper (TFH) cell precursors. Here we show that, upon treatment withdrawal, these Foxp3 TR1 cells spontaneously differentiate into a novel immunoregulatory Foxp3 TR1 subset that inherits epigenetic and transcriptional hallmarks of their precursors, including clonotypic T-cell receptors, and is distinct from other Foxp3 Treg subsets.

High-grade serous ovarian carcinoma (HGSOC) is one of the deadliest malignancies in female population and the cause of 70% of all ovarian cancer-related deaths. Among its hallmarks, the fluid accumulation in the peritoneal cavity, or ascites, is a peculiar pathological sign during late stages and in recurrent patients. Besides cancer cells, ascitic fluids contain a heterogeneous cellular composition, representing a precious source to dissect molecular mechanisms underlying invasion and metastatization or find new biomarkers to predict therapy response.

Menopause, an aged process due to loss of permanent ovarian function (mainly decreased production of estrogen) without another pathological or physiological cause becomes one of the most health issues in the world. Vasomotor symptoms (VMS) is a hallmark of menopause, occurring in up to 80 % of women experiencing and persisting for over seven years with significant negative impacts on physical, psychological, social, and overall wellbeing. Conventionally, menopause hormone therapy (MHT) is considered the most effective therapeutic opinion for many years based on the fact as "deficiency of estrogen"; however, the ratio of benefits and risk is highly debated and always in concern.

Mitochondrial dysfunction is a hallmark of woody breast myopathy in broiler chickens.

The woody breast (WB) myopathy poses significant economic and welfare concerns to the poultry industry, however, there is no effective strategy to mitigate this pathology due to its unknown etiology. After showing previously that hypoxia is a key factor in WB progression, we used here various techniques demonstrating dysregulated mitochondria (morphology, biogenesis, tethering, function, and bioenergetics) in WB-affected muscles and in hypoxic myoblasts compared to healthy tissues and normoxic cells, respectively. The increased levels of calcium (Ca) in both WB-affected tissues and hypoxic myoblasts suggested that mitochondrial Ca overload is likely a leading cause for mitochondrial dysfunction that merits further in-depth investigation.
