The administration of therapeutics using bioconjugation has been mainly limited to drugs containing amine, alcohol, or thiol functional groups. Here, we report a general procedure for the preparation of benzylic N-acyl carbamates suitable for masking the amide group in important drugs such as Linezolid, Enzalutamide, or Tasimelteon in good to acceptable yields. These N-acyl carbamates appear to be stable in plasma, while a qualitative analysis of further drug uncage demonstrates that, at pH values of 5.5, a classical 1,6-benzyl elimination mechanism takes place, releasing more than 80% of the drug in 24 h.

We report on the syntheses of NeuAc and NeuGc-containing glycosides via the use of double carbonyl-protected N-acetyl sialyl donors. The 7-O,9-O-carbonyl protection of an N-acyl-5-N,4-O-carbonyl-protected sialyl donor markedly increased the α-selectivity during glycosylation, particularly when glycosylating the C-8 hydroxyl group of sialic acids. The N-acyl carbamates were selectively opened with ethanethiol under basic conditions to provide N-acyl amines.

In order to investigate the influence of a minor variation in structure of N-acyl chitosan derivatives on enantioseparation, chiral selectors (CSs) of chitosan 3,6-bis(phenylcarbamate)-2-(phenylacetamide)s and chitosan 3,6-bis(phenylcarbamate)-2-(cyclohexylacetamide)s were synthesized. The corresponding chiral stationary phases (N-PhAc CSPs and N-cHeAc CSPs) were also prepared, respectively, with the two series of CSs. Enantioseparation results revealed that the N-PhAc CSPs were better than the N-cHeAc ones in enantioseparation.

We developed an electrochemical carboamidation sequence that affords either cyclic β-amidoamine products via direct functionalization or linear hydroxybisamide products via a ring opening pathway. The reaction pathway was dependent on the nature of the -acyl activating group, with carbamate groups favoring direct isocyanide addition to the -acyliminium ion intermediate and the benzoyl activating group favoring the ring opening-functionalization pathway. Both protocols are one-pot reaction sequences, have general applicability, and lead to peptide-like products of greatly increased molecular complexity.

We present a systematic investigation on an improved variant of the -acyl-Pictet-Spengler condensation for the synthesis of 1-benzyltetrahydroisoquinolines, based on our recently published synthesis of -methylcoclaurine, exemplified by the total syntheses of 10 alkaloids in racemic form. Major advantages are a) using ω-methoxystyrenes as convenient alternatives to arylacetaldehydes, and b) using the ethoxycarbonyl residue for both activating the arylethylamine precursors for the cyclization reaction, and, as a significant extension, also as protective group for phenolic residues. After ring closure, the ethoxycarbonyl-protected phenols are deprotected simultaneously with the further processing of the carbamate group, either following route A (lithium alanate reduction) to give -methylated phenolic products, or following route B (treatment with excess methyllithium) to give the corresponding alkaloids with free N-H function.
