Cytotoxicity assays are essential tests in studies on the safety and biocompatibility of various substances and on the efficiency of anticancer drugs. The most frequently used assays commonly require application of externally added labels and read only collective response of cells. Recent studies show that the internal biophysical parameters of cells can be associated with the cellular damage. Therefore, using atomic force microscopy, we assessed the changes in the viscoelastic parameters of cells treated with eight different common cytotoxic agents to gain a more systematic view of the occurring mechanical changes. With the robust statistical analysis to account for both the cell-level variability and the experimental reproducibility, we have found that cell softening is a common response after each treatment. More precisely, the combined changes in the viscoelastic parameters of power-law rheology model led to a significant decrease of the apparent elastic modulus. The comparison with the morphological parameters (cytoskeleton and cell shape) demonstrated a higher sensitivity of the mechanical parameters versus the morphological ones. The obtained results support the idea of cell mechanics-based cytotoxicity tests and suggest a common way of a cell responding to damaging actions by softening.

The firmness of the two apple varieties: Idared and Pinova was similar during ripening, while it decreased significantly during 3-month storage only for Idared. Pectin-rich fractions were isolated from apple flesh tissue: water-soluble pectin (WSP), imidazole-soluble pectin (ISP), and hemicellulose-rich fractions: natively acetylated hemicelluloses (LiCl-DMSO), deacetylated hemicelluloses (KOH). It was shown that the degree of acetylation (DAc) of the hemicelluloses fraction (LiCl-DMSO) increased during apple ripening and storage, with higher values for Idared.

Quantifying the mechanical response of the biological milieu (such as the cell's interior) and complex fluids (such as biomolecular condensates) would enable a better understanding of cellular differentiation and aging and accelerate drug discovery. Here we present time-shared optical tweezer microrheology to determine the frequency- and age-dependent viscoelastic properties of biological materials. Our approach involves splitting a single laser beam into two near-instantaneous time-shared optical traps to carry out simultaneous force and displacement measurements and quantify the mechanical properties ranging from millipascals to kilopascals across five decades of frequency.

Abscisic acid (ABA) is a key hormone in plant growth and development, playing a central role in responses to various biotic and abiotic stresses as well as in fruit ripening. The present study examined the impact of ABA and nordihydroguaiaretic acid (NDGA) on various postharvest 'Docteur Jules Guyot' pear fruit characteristics, including firmness, pectinase activity, pectin content, volatile aromatic substances, and the expression of correlated genes. The results showed that ABA quickly reduced fruit firmness, increasing the activity of pectin degradation-related enzymes.

Laminating a free-standing carbon electrode film onto perovskite film is a promising method for fabricating HTM (hole transport material)-free carbon electrode perovskite solar cells (c-PSCs), offering more flexibility by decoupling the processes of carbon electrode and perovskite layer formation. However, the power conversion efficiency (PCE) of laminated HTM-free c-PSCs (<16.5%) remains lower compared to c-PSCs with printed carbon pastes (>20%), primarily due to poor interfacial contact between the perovskite and carbon layers.

Texture is an important quality index reflecting a series of sensory evaluations of fruit, and softening is the main texture change of most postharvest fruit. Persimmon fruit () shows a pattern of decreasing firmness after harvest, leading to a short duration of sale and excessive waste. We found that the treatment with gibberellin (GA) could effectively inhibit the postharvest softening of persimmon fruit, but the underlying mechanism is unclear.
